摘  要:为研究花色苷生物合成途径中类黄酮3′羟化酶基因(F3′H)的序列信息,以泰山红石榴花瓣为试材,提取总RNA后反转录为cDNA,根据GenBank中登录的相关物种F3′H基因的保守序列设计兼并引物,对反转录后的cDNA进行PCR扩增并测序。结果表明:在石榴中克隆到了长度为996bp的F3′H基因cDNA片段,该基因编码331个氨基酸,含有细胞色素P450保守域,属于CYP75B亚家族。该序列在GenBank中的登录号为KC430328。The flower of pomegranate cultivar Taishanhong was used as experimental materials. Total RNA was extracted and reversely transcribed into cDNA. Degenerate primers were designed according to conserved sequences of flavonoid 3Chydroxylase (F3tH) gene of some related species in GenBank database. PCR amplication was conducted and then the PCR product was sequenced. The result showed that a F3'H cDNA fragment was cloned from pomegranate flower with the sequence length of 996 bp,coding 331 amino acids. The F3'H in pomegranate contained eytochrome P450 conserved domains,belonging to CYP75B subfamily. The accession number for the gene in GenBank was KC430328.
