两种培养方法检测解脲脲原体及其菌落形态学的比较研究

摘　　要：目的比较选择性固体培养基与液体培养基检测男女不同类型生殖泌尿道标本中解脲脲原体(Uu)结果的差异,并观察Uu在固体培养基上的菌落形态学特征。方法 1 698例生殖泌尿道标本被分别接种于固体培养基和液体培养基中,比较不同类型标本中Uu的阳性率、2种培养基的阳性率以及菌落形态学的差异。结果女性宫颈分泌物标本中Uu阳性率(49.9%)显著性高于男性精液和尿道分泌物标本(39.6%、19.2%,P均<0.05);液体培养法与固体培养法相比,女性宫颈分泌物标本Uu阳性率差异有统计学意义(P<0.05),而男性精液和尿道分泌物标本Uu阳性率之间差异无统计学意义(P>0.05);Uu菌落形态以海胆型为主(77.5%),其次是油煎蛋样(13.8%),空泡型、微小型、棒状和混合型较少见。结论女性宫颈分泌物标本中Uu阳性率最高,男性尿道分泌物标本Uu阳性率最低。对于女性宫颈分泌物标本,2种方法检测Uu的结果差异较大;对于男性尿道分泌物标本,2种方法检测结果一致性较好。Uu在菌落形态学方面存在多种异质性。Objective To determine Ureaplasma urealyticum（Uu） from different kinds of samples from males and females by the solid medium culture and the liquid medium culture, and to observe the morphological characteristics of Uu colonies on the solid medium culture. Methods The 1 698 samples from urogenital tract were cultured in the solid medium culture and the liquid medium culture. The positive rates from the 2 kinds of assays and different kinds of samples were compared and analyzed. The colony morphological heterogeneity was analyzed. Results The positive rate of Uu from female cervix（49.9% ） was significantly higher than those from male semen and urethral secretion（39.6% , 19.2% , P 〈 0.05 ）. There was statistical significance of the positive rate from female cervix between the liquid medium culture and the solid medium culture（ P 〈 0.05 ）. However, there was no statistical significance of the positive rate from male semen and urethral secretion between the 2 kinds of assays （ P 〉 0.05 ）. The main colonies of Uu was typical echinus colony（77.5% ） , followed by fried egg colony（ 13.8% ）, vesicle colony, tiny colony, coryneform colony and mixed colony. Conclusions The positive rate of Uu from female cervix is the highest. The positive rate from male urethral secretion is the lowest. The results of Uu from female cervix by the 2 kinds of assays are different obviously. There is a good consistency between the 2 kinds of assays for male urethral secretion. Uu colonies have morphological heterogeneity.
