摘  要:目的:观察豚鼠血清中白介素-4(IL-4)、γ-干扰素(IFN-γ)含量以及T-bet和GATA-3 mRNA的表达情况,探讨穴位敷贴对过敏性哮喘的免疫调节机制。方法:将30只雄性豚鼠随机分为正常组、模型组、穴位组、非穴位组、阳性对照组(氨茶碱组),每组6只。以卵清白蛋白致敏激发法建立哮喘模型,采用ELISA法检测豚鼠血清中IL-4、IFN-γ含量,RT-PCR法检测肺组织T-bet和GATA-3 mRNA的表达。结果:穴位组与非穴位组IL-4血清含量、GATA-3 mRNA表达明显低于模型组,IFN-γ含量、T-bet mRNA表达明显高于模型组(P<0.01),非穴位组血清IL-4、IFN-γ含量,T-bet mRNA和GATA-3 mRNA的表达与穴位组相比有显著性差异(P<0.01)。结论:穴位敷贴可以通过调节转录因子的表达,从而改变Th0细胞分化方向,影响特征性细胞因子的分泌,纠正Th1/Th2比例失衡,这是穴位敷贴控制哮喘气道炎症的一种可能的机制,且药物敷贴于穴位处较之非穴位处能够发挥更好的疗效。Objective: To observe guinea pig serum IL-4, IFN-y content and T-bet and GATA-3 mRNA expression, to discuss the immune regulation mechanism of acupoint application to allergic asthma. Methods: Thirty guinea pigs were randomly divided into normal group, model group, acupoint application group, non-acupoint application group and positive control group (aminophylline group), six guinea pigs in each group. The asthmatic model was established by immunization with intraperitoneally injected and inhaled ovalbumin (OVA). Enzyme-linked immunosorbent assay (ELISA) was used to detect the levels of IL-4 and IFN-y, and reverse transcription polymerase chain reaction (RT-PCR) assay was used for the detection of mRNA expression of T-bet and GATA-3 mRNA. Results: IL-4 content and GATA-3 expressions in the acupoint group and the non-acupuncture point group were significantly lower than the asthma group; IFN-y content and T-bet expressions were obviously higher than those of the asthma group (P〈0.01). IL-4 and IFN-7 contents, T-bet and GATA-3 mRNA expressions in the acupoint group had significant differences from the non-acupoint group (P〈 0.01). Conclusion: Acupoint application can change Th0 cell differentiation direction, influence IL-4/IFN-y secretion, correct Thl/Th2 imbalance through regulation of the transcription factors T-bet/GATA- 3 expression which may be one of possible mechanisms acupoint application control asthmatic airway inflammation. Application in acupoint places can play a better curative effect than in non-acupoint places, which may relate to the physiological effects of special points, and this can be a new starting point for further study on acupoint application effect mechanisms.
