摘  要:目的:观察糖肾平含药血清对高糖环境下脂多糖(LPS)刺激足细胞增殖与凋亡的影响。方法:本实验在前期整体动物实验的基础上,进行足细胞传代培养后,将细胞种植于96孔培养板,分为正常组,高糖组,高糖+LPS组,厄贝沙坦组,糖肾平小、中、大剂量组及抑制剂组,分别在含药血清作用12、24、36、48h时,用MTT法检测各组足细胞增殖与凋亡情况。结果:高糖环境中足细胞经LPS刺激24h时增殖明显,糖肾平不同剂量组能明显抑制高糖环境中LPS刺激的足细胞增殖,与高糖组和高糖+LPS组相比,有显著性差异(P<0.01);高糖组和高糖+LPS刺激36h时足细胞开始凋亡,48h时出现较多凋亡,糖肾平不同剂量组能明显抑制足细胞凋亡,与高糖组和高糖+LPS组相比,糖肾平大、中、小剂量组有显著性差异(P<0.01,P<0.05)。结论:糖肾平对高糖+LPS刺激足细胞的增殖和凋亡有一定的调节作用。Objective: To observe the effect of Tangshenping medicated serum on the proliferation and apoptosis of glomerular epithelial cells (podocytes) in a high glucose and LPS environment. Methods: On the basis of the previous in vivo animal experiment, the podocytes were cultivated and planted on the 96-well fiat bottom plate. The ceils were divided into the following eight groups: control group, high glucose group, high glucose with LPS group, Irbesartan group, low dose Tangshenping group, medium dose Tangshenping group, high dose Tangshenping group, inhibitor group. MTT assay was used to measure the proliferation and apoptosis of podocytes at 24, 36, and 48 hour time intervals. Results: Proliferation of podocytes in high glucose and LPS conditions was obvious at the 24-hour mark, while proliferation was significantly inhibited (P〈0.01) in the Tangshenping groups compared to the high glucose and high glucose with LPS groups. The podocytes began to apoptosis at 36-hour time interval and became apparent at 48-hours, but it was hindered in the Tangshenping groups. Compared with the high glucose group and high glucose with LPS group, low and medium and high doses of Tangshenping yielded significant differences (P〈0.01, P〈0.05). Conclusion: Tangshenping has a definitive regulatory effect on the proliferation and apoptosis of podocytes in a high glucose and LPS environment.
