摘 要:目的探讨急性应激抑郁模型大鼠血浆脑源性神经营养因子的水平及外周粒细胞与脑组织中脑源性神经营养因子的表达。方法将16只成年雄性大鼠随机分为实验组与对照组,每组8只,实验组大鼠连续接受电击诱导3d,采用旷场试验评价两组大鼠行为,安乐处死,取两组血样与脑组织(海马、前额叶、中缝核及纹状体),采用双抗体夹心ABC—ELISA法和实时荧光定量PCR(Real-TimePCR)检测法检测血浆脑源性神经营养因子水平及外周粒细胞与各脑区脑源性神经营养因子的表达。结果实验组大鼠连续接受电击诱导3d后,行为明显减少,越线次数、头动总距离显著低于对照组(P〈0.05或0.01);血浆脑源性神经营养因子水平及脑组织中脑源性神经营养因子表达与对照组比较差异均无显著性(P〉0.05),外周粒细胞脑源性神经营养因子的表达显著低于对照组(P〈0.01)。结论急性应激不引起大鼠血浆脑源性神经营养因子水平及脑组织中脑源性神经营养因子表达的改变,但大鼠外周粒细胞脑源性神经营养因子的表达降低。Objective To explore the levels of plasma brain-derived neurotrophic factor (BDNF) and BD- NF expression in peripheral granulocyte and brain tissue of acute stress depression model rats. Methods Sixteen SD rats were randomly assigned to experimental and control group (both n=8), continuous dec- trostimulation inductions were given to rats of experimental group for 3 days, rats' behaviors of both groups evaluated with the Open Field Test, they put to death through peace and happiness, blood samples and brain tissues (hippocampus, prefrontal lobe, rapheal nuclei and corpus striatum) taken, levels of plas- ma BDNF and BDNF expression in peripheral granulocyte and each encephalie region detected by Double Antibody Sandwich ABC-ELISA method and Real-Time PCR. Results After 3-day continuous electros- timulation inductions behaviors obviously reduced, over-line frequency and total distance of head movement were significantly lower in rats of experimental group compared with control group (P〈0.05 or 0.01); there were no significant differences in levels of plasma BDNF and BDNF expression in brain tissues (P〈 0.05), BDNF expression in peripheral granulocyte was significantly lower in experimental than control group (P〈0.01). Conclusion Acute stress does not alter the level of plasma BDNF and BDNF expression of brain tissue, but BDNF expression of rats peripheral granulocyte reduces.
