摘  要:以4个番茄品种为材料,采用改进的氮蓝四唑光还原法和聚丙烯酰胺凝胶电泳法,研究了经聚乙二醇模拟干旱胁迫处理后,番茄幼苗叶片超氧化物歧化酶(SOD)同工酶活性及酶谱的变化。结果表明,4个番茄品种的SOD同工酶活性均较对照明显增加,各品种的胁迫系数高低顺序依次为中蔬4号、中杂105、Money maker、中杂9号;模拟干旱胁迫处理没有使各品种SOD同工酶酶谱条带数目出现增加,仍为3条,SOD同工酶条带对干旱响应敏感程度的大小排序依次为SOD-Ⅲ、SOD-Ⅱ、SOD-Ⅰ,其中SOD-Ⅲ条带的酶谱亮度、带宽在处理与对照之间均明显增加,增加程度大小的品种排序依次为中蔬4号、中杂105、Money maker、中杂9号,与胁迫系数一致,说明SOD与干旱胁迫密切相关的同工酶为SOD-Ⅲ,其可能是番茄耐旱品种SOD同工酶活性较高的根源;所以SOD-Ⅲ条带可作为番茄抗旱生理育种的筛选指标。Using 4 tomato(Solanum lycopersicum L.) cultivars as materials,with improved nitrogen and light blue tetrazolium reduction method and polyacrylamide gel electrophoresis,the dynamics of superoxide dismutase(SOD) isozyme activity and zymogram in leaves of tomato seedlings under poly ethylene glycol(PEG) simulated drought stress were studied.Results showed that SOD isozyme activity of 4 tomato cultivars were significantly increased compared with control.The stress coefficient ranked from high to low as S.lycopersicum cv.Zhongshu No.4,S.lycopersicum cv.Zhongza 105,S.lycopersicum cv.Moneymaker,S.lycopersicum cv.Zhongza No.9.The number of SOD isozyme zymogram bands was 3 under simulated drought stress treatment,which was not increased.The sensitivity of SOD isozymes bands to drought response ordered from high to low as SOD-Ⅲ,SOD-Ⅱ,SOD-Ⅰ.The brightness and width of SOD-Ⅲ enzymes band was significantly increased compared with control,while the increase range ranked from high to low was S.lycopersicum cv.Zhongshu No.4,S.lycopersicum cv.Zhongza 105,S.lycopersicum cv.Moneymaker,S.lycopersicum cv.Zhongza No.9,which was consistent with the stress coefficient,indicating that SOD-Ⅲ was closely related to drought stress,thus could be the reason why drought-tolerant tomato varieties had higher SOD isozyme activity.SOD-III could be used as indicator for breeding of drought-resistant tomato.
