摘 要:目的:探讨归芪多糖对免疫低下小鼠迟发型超敏反应(DTH)的影响。方法:将40只昆明种小鼠随机分为正常对照组、模型对照组、归芪多糖高剂量组(100 mg/kg)、归芪多糖低剂量组(50 mg/kg)和阳性药物对照组[黄芪精口服液(1.8 g/kg)],每组8只。除正常对照组外其余各组通过皮下注射环磷酰胺(CY)建立免疫抑制小鼠模型。模型对照组灌服等体积生理盐水,各药物组分别灌服归芪多糖(50 mg/kg和100 mg/kg)和黄芪精口服液(1.8 g/kg),1次/d,连续灌服6天后,除正常对照组外其余各组以二硝基氟苯(DNFB)致敏并加强2天后计算小鼠耳廓肿胀度及脾脏、胸腺指数。结果:100 mg/kg归芪多糖能明显增加免疫抑制小鼠耳廓肿胀度,与阳性对照药物黄芪精口服液相比无显著性差异;且归芪多糖在50~100mg/kg剂量范围内可对抗环磷酰胺所致小鼠脾脏和胸腺的萎缩,增加小鼠免疫脏器指数。结论:归芪多糖可恢复小鼠对外源性过敏原所导致的迟发性超敏反应,且能对抗环磷酰胺引起的脾脏和胸腺的萎缩。Objective: To explore the effects of angelica and astragalus polysaccharide on delayed type hypersensitivity (DTH) of immunosuppressive mice. Method: All 40 mice were randomized into normal control group, model control group, high (100 mg/kg) and low (50 mg/kg) dosage groups of angelica and astragalus polysaccharide, and the control group of positive drug (HuangQiJing oral liquid, 1.8 g/kg), eight mice each group. Except normal control group, immunosuppressive mice models were established with hypodermic injection of cyclophosphamide (CY). Normal control group received the same amount of physiological saline and other groups were given with angelica and astragalus polysaccharide(100 mg/kg and 50 mg/kg) and HuangQiJing oral liquid (1.8 g/kg), once each day, for six consecutive days, except normal control group, other groups were sensitized with dinitrofluorobenzene (DNFB) and enhanced for two days to calculate the swollen degree of auricle, the indexes of spleen and thymus. Result: Compared with HuangQiJing oral liquid, 100mg/kg angelica and astragalus polysaccha- ride enhanced the swollen degree of auricle and showed no significant difference; angelica and astragalus polysaccharide could resist the atrophy of spleen and thymus in mice induced by CY and enhance the index of immune organs in mice. Conclusion: Angelica and astragalus polysaccharide could recover delayed type hypersensi- tivity and resist the atrophy of the spleen and thymus induced by CY.
