摘 要:为了建立"清瘟败毒颗粒"中盐酸小檗碱含量的高效液相色谱检测方法(high-per-formance liquid chromatography,HPLC),采用SB-C18柱(StableBond,4.6×150 mm,5μm),流动相:乙腈-0.05mol/L磷酸二氢钾溶液(50∶50),紫外检测波长为345nm,流速1.0mL/min,柱温30℃。在选择色谱条件下,盐酸小檗碱与辅料、溶剂及清瘟败毒颗粒中的其他成分的峰之间分离良好,在16.0~48.0μg/mL范围内峰面积与盐酸小檗碱含量直线关系(r2=1,n=6),平均回收率为99.09%。该方法简便、灵敏、可重复性好,可用于清瘟败毒颗粒的质量控制。To establish a simple and sensitive HPLC method for determining berberine hydrochloride in Qingwen Baidu granule,HPLC method was used. The HPLC system consisted of Stable Bond C18 column (4.6×150mm,5μm) and a mobile phase of acetonitrile-0.05mol/L potassium dihydrogen phosphate solution (50 : 50, v : v) mixture. Berberine hydrochloride was detected at 345 nm, with a flow rate of 1.0 mL/min. The column temperature was maintained at 30 ℃. The excipients and solvents in the granule could be well separated from the drug under such a designated chromatogram condition and did not interfere with the assay. A good linear relationship was found between peak area and the concentration of berberine hydrochloride in the range of 16.0-48.0μg/mL. The average recovery of berberine Hydrochloride was 99.09%. The established approach was specific, accurate, reliable, prompt, sensitive and applicable. It could be used to control the quality of Qingwen Baidu granule.
