庆大霉素对豚鼠血管纹Na,K-ATP酶α亚单位异构体表达的影响

摘　　要：目的　观察正常豚鼠血管纹Na ,K -ATP酶α亚单位异构体的表达及庆大霉素对其表达的影响。方法　用鼠抗大鼠Na ,K -ATP酶α亚单位异构体特异性单克隆抗体 ,采用免疫组化SP法观察正常豚鼠血管纹中Na,K -ATP酶α1 异构体的表达及庆大霉素作用后表达的变化。结果　正常豚鼠血管纹Na,K -ATP酶α1 异构体表达呈阳性 ,而α2 和α3 异构体表达呈阴性。庆大霉素作用后α1 异构体表达显著减弱。结论　庆大霉素显著抑制豚鼠血管纹Na ,K -ATP酶α异构体的表达 ,从而抑制血管纹Na ,KObjective To investigate the expression of a subunit isoforms of Na,K-ATPase in stria vascularis of guinea pig,and the effect of gentamicin on it.Methods The expression of a subunit isoforms of Na,K-ATPase in stria vascualris of guinea pig and its change after gentamicin exposure were identified immunocytochemically using the specific mouse antibodies against the α 1,α 2 and α 3 subunits of rat Na,K-ATPase.Results All marginal cells,intermediate cells and basal cells of stria vascualris showed positive expression of α 1 isoform and negative expression of α 2 and α 3 isoforms.The immunostaining of α 1 isoform was attenuated significantly after gentamicin esposure.Conclusion Gentamicin significantly inhibits the expression of Na,K-ATPase α 1 subunit isoform of the stria vascularis ,sequentially inhibits the activity of Na,K-ATPase.
