SOCS3蛋白与IL-13对促进人平滑肌细胞增殖作用的相关研究

摘　　要：目的探讨人平滑肌细胞系中细胞因子信号抑制因子3(suppressor of cytokine signaling 3,SOCS3)蛋白的表达及其在IL-13促细胞增殖中的作用机制。方法应用RT-PCR方法测定IL-13作用前后细胞系中SOCS3的表达情况,用MTT比色法测定HSMC增殖情况。结果 RT-PCR法显示,IL-13作用后,SOCS3的表达明显升高;MTT比色法结果显示,IL-13作用后细胞增殖情况明显(P<0.01)。结论 IL-13能促进HSMC的增殖,其可能机制为IL-13刺激HSMC细胞分泌SOCS3蛋白增多,并活化JAK/STAT途径,引起细胞增殖。Objective To investigate cytokine signaling inhibition factor 3 （SOCS3） protein expression in human smooth muscle cell line and its mechanism to promote cell proliferation in IL-13. Methods SOCS3 expression was detected by RT-PCR method before and after the IL-13 acted on cell lines, the proliferation of HSMC was detected by MTF assay. Results The result of RT-PCR showed that SOCS3 expression was significantly increased with stimulation of IL-13; the results of MTF assay was the same （P 〈0. 01）. Conclusion IL-13 can promote the proliferation of HSMC, the mechanism might be that IL-13 can increase the secretion of SOCS3, and activate the pathway of JAK/STAT which cause the proliferation of cells.
