摘  要:目的:初步探讨腺苷A2A受体在过敏性哮喘疾病中的表达水平变化和在哮喘发生、发展过程中的作用。方法:分别选择18例对屋尘螨过敏的慢性持续期哮喘患者(过敏性哮喘组)和19名健康志愿者(正常对照组),通过荧光实时定量PCR方法检测研究对象外周血单个核细胞中A2A受体mRNA水平。酶联免疫吸附试验(ELISA)检测血浆中屋尘螨特异性免疫球蛋白(Ig)E、白细胞介素(IL)-4、IL-17A和转化生长因子(TGF)-β水平。结果:过敏性哮喘组A2A受体mRNA明显高于正常对照组[(0.45±0.29)×10-3比(0.21±0.06)×10-3;t=3.611,P<0.01],且A2A受体mRNA与血浆IL-4水平呈正相关(r=0.583,P<0.01),与TGF-β水平呈负相关(r=-0.499,P<0.01),与IL-17A无相关性(r=0.395,P=0.085)。结论:腺苷A2A受体可能参与了过敏性哮喘的发生、发展。Objective To investigate the expression of adenosine A2A receptor(A2AR) and its role in the development and progress of allergic asthma.Methods Eighteen patients with persistent asthma allergic to house dust mite and 19 healthy volunteers served as control were enrolled.The expression of A2AR mRNA in peripheral blood mononuclear cells(PBMCs) was determined by SYBR Green Ⅰ relative quantitative real-time PCR.The level of specific house dust mites IgE,interleukin(IL-4),IL-17A and transforming growth factor(TGF)-β were detected by ELISA.Results Expression of A2AR mRNA was significantly higher in patients with allergic asthma than that in normal controls [(0.45 ± 0.29)×10-3 vs(0.21±0.06)×10-3,t=3.611,P0.01].A2AR mRNA was positively correlated with IL-4(r=0.583,P0.01),negatively correlated with TGF-β(r=0.499,P 0.01),and no correlation was found with IL-17A(r =0.395,P =0.085).Conclusions A2AR might participate in the development and progress of allergic asthma.
