摘 要:从中华大蟾蜍(Bufo gargarizans)某一个体的皮肤第一链cDNA中克隆到多个编码半乳糖凝聚素3(Gal-3)的cDNA。为分析该现象在中华大蟾蜍中的普遍性,分离并纯化3个不同个体的皮肤总RNA并合成其第一链cDNA,然后以此为模板,通过DNA聚合酶链式反应(PCR)扩增gal-3的开放阅读框序列。序列分析结果表明,从3个不同个体中均克隆到多个不同的gal-3 cDNA的ORF,在3组不同的试验中,出现完全相同的克隆或者完全相同的缺失以及相同的多核苷酸多态性位点,支持gal-3 cDNA多样性的客观存在,揭示gal-3cDNA的多样性是该物种的特征。The skin galectin-3 (gal-3) cDNA diversity of Bufo gargarizans was discovered encoding Galectin-3 proteins ( Gal-3 ). In order to know if this phenomenon is a common event in this species, three independent skin first strand cDNAs were synthesized from the total RNA of three different B. gargarizans individuals, which were used as templates for subsequent polymerase chain reaction (PCR) amplification of gal- 3 open reading frame (ORF), and the PCR products were cloned into pGM-T vector and sequenced. As the result, gal-3 cDNA diversity in- cluding high frequency of SNP ( single nucleotide polymorphism) and several deletions were found in all three independent experiments based on the three different individuals. However, this cDNA diversity did not result in the important amino acid absence or substitution of Gal-3, such as phosphorelation sites and galectoside binding sites. These data suggests that the gal-3 cDNA diversity is a common event of this spe- cies, which might capacitate B. gargarizans individuals strong adaptative potential to everchangeful habitats.
