双夹心DIGFA检测血吸虫循环抗原的应用研究

摘　　要：以日本血吸虫重组极低密度脂蛋白 ( SVL BP)制备的多抗为捕捉抗体 ,胶体金标记血吸虫虫卵多克隆抗体为覆盖抗体 ,采用双夹心金标免疫渗滤法 ( DIGFA)检测血吸虫病病人血清中循环抗原。结果 70份急性血吸虫病病人血清的阳性率为 10 0 % ,15 9份慢性血吸虫病病人血清的阳性检出率为 6 2 .3% ;12 0份流行区健康人群血清检测均阴性 ;10 0份非流行区健康人群血清 ,2份呈阳性反应 ,假阳性率为 2 %。与肺吸虫病病人血清的交叉反应率为4.0 % ( 1/ 2 5 ) ;与华支睾吸虫病病人血清未见交叉反应。提示该法具有较高的敏感性和特异性 ,判断结果比较客观 ,且简便、快速 。To use recombinant proteinSVLBP as diagnostic reagent and rabbit antiSEA PcAb as covering antibody, circulating antigen of Schistosoma japonicum in patients sera was detected by sandwich dot immunogold filtration assay(DIGFA). The results showed that the positive rates of patients with acute schistosomiasis and patients with chronic schistosomiasis were 100%(70/70) and 62.3%(99/159) respectively. All of 120 sera from normal population in epidemic area were negative, while 2 of 100 sera from nonepidemic area were positive. The rate of cross reaction with the sera of patients with paragonimiasis was 4.0%(1/25), there was no cross reaction with sera from patients with clonorchiasis. Sandwich DIGFA was not only easier to operate and rapid, but also sensitive and specific for detection of circulating antigens of schistosome. It is suggested that sandwich DIGFA is suitable for diagnosis of schistosomiasis in the basic units and field.
