摘 要:目的:探讨人脐带间充质干细胞(human umbilical cord mesenchymal stem cells,HUMSCs)在脂多糖(Lipopolysaccharide,LPS)诱导的新生SD大鼠急性肺损伤的作用。方法:人脐带间充质干细胞的分离、培养和鉴定。将30只新生SD大鼠随机分成生理盐水(NS)对照组、LPS模型组和HUCMSCs移植组,每组10只。显微镜观察各组大鼠肺组织病理变化,ELISA检测血清中炎性细胞因子的表达差异。结果:与对照组相比,LPS模型组肺湿/干重比(W/D)和肺组织髓过氧化物酶(MPO)活性升高,血清中TNF-α和IL-6的表达显著增加(P<0.05)。与LPS模型组相比,HUCMSCs移植组W/D和MPO活性有所下降,血清中TNF-α和IL-6的表达明显降低(P<0.05)。结论:在LPS诱导的新生大鼠肺损伤模型中,人脐带间充质干细胞移植能够减轻肺组织损伤,并抑制炎性因子的表达。Objective. To explore the effects of human umbilical cord mesenchymal stem cells (HUMSCs) in lipopolysaccharide induced acute lung injury in neonatal rats. Methods. HUMSCs were iso- lated, cultured and identified. Thirty neonatal SD rats were divided into control group, LPS group and HUMSCs transplantation group randomly (n=10). Pathological changes in lung were observed under mi- croscopy. The level of inflammation factors in rats sera were detected by ELISA. Results. Compare with the control group, lung W/D ratio, MPO activity, TNF-α and IL-6 in sera were significantly increased in LPS group (P〈0.05). Compare with LPS group, lung W/D ratio , MPO activity, TNF-α and IL-6 level in HUMSCs transplantation group were decreased (P〈0.05). Conclusion. HUMSCs can alleviate acute lung injury induced by lipopolysaccharide and decrease the sera levels of inflammation factors in neonatalrats.
