摘 要:目的观察糖络宁对链脲佐菌素(STZ)诱导糖尿病大鼠一般指标及摆尾温度阈值的影响,探讨其对糖尿病性周围神经病变(DPN)的防治作用。方法采用一次性腹腔内注射60 mg/kg STZ诱导糖尿病大鼠模型。将模型大鼠随机分为模型组、糖络宁组、α-硫辛酸组,每组23只,另取15只大鼠为正常组。糖络宁组予糖络宁20 g/(kg.d)灌胃,α-硫辛酸组予α-硫辛酸20 mg/(kg.d)灌胃,正常组和模型组大鼠予等体积蒸馏水灌胃,连续处理12周。观察大鼠体质量(BM)、空腹血糖(FPG)、糖化血红蛋白(HbA1c)及摆尾温度阈值的变化。结果与正常组比较,各组糖尿病大鼠BM明显下降(P<0.01),而FPG、HbA1c及摆尾温度阈值均明显升高(P<0.01);经糖络宁、α-硫辛酸处理后,2组大鼠FPG、HbA1c及摆尾温度阈值均明显改善(P<0.05),但BM无明显变化。糖络宁组大鼠在FPG改善方面与α-硫辛酸组比较差异有统计学意义(P<0.05),其余指标差异均无统计学意义(P>0.05)。结论糖络宁能有效改善糖尿病大鼠的糖毒性和摆尾温度阈值,对DPN有防治作用。Objective To observe the effects of Tangluoning on general indices and tailflick threshold temperature(TTT) of streptozocin(STZ) induced diabetic rats.Methods Diabetes mellitus was induced by a single intraperitoneal injection 60 mg/kg STZ in male SD rats.Sixty-nine diabetic rats were randomly assigned to the model group,Tangluoning group and alpha lipoic acid(ALA) group(23 rats in each group).Fifteen normal rats were randomly set as normal group.Tangluoning group was performed intragastric administration of 20 g/(kg.d) crude drug of Tangluoning.ALA group was administrated with 20 mg/(kg.d) ALA.Normal group and model group were administrated with distilled water of equal volume.All groups were treated for twelve weeks.The changes of body mass(BM),fast plasma glucose(FPG),glycolated hemoglobin(HbA1c) and tailflick threshold temperature(TTT) of rats were measured.Results Compared with normal group,the levels of FPG,HbA1c and TTT of all diabetic rats were significantly increased(P 0.01),and the levels of BM were significantly reduced(P 0.01).Compared with the model group,the levels of FPG,HbA1c and TTT of rats in both Tangluoning group and ALA group were significantly improved(P 0.05),but BM had no significant change(P 0.05).The effect of Tangluoning group on FPG was superior to that of ALA group,the difference was significant(P 0.05),but effects on BM,HbA1c and TTT had no significant difference(P 0.05).Conclusion Tangluoning can effectively improve glucose toxicity and TTT of diabetic rats.Tangluoning has preventive and therapeutic effect on diabetic peripheral neuropathy.
