活血化瘀通络中药对糖尿病胃轻瘫大鼠胃排空及胃促生长素的影响

摘　　要：目的研究活血化瘀通络中药对糖尿病胃轻瘫大鼠胃排空及胃促生长素的影响。方法健康雄性SD大鼠50只,按体质量随机分为对照组、模型组、中药1组(丹参、川芎)、中药2组(水蛭、地龙)、中药3组(丹参、川芎、水蛭、地龙)。除对照组外,其余各组大鼠腹腔注射链脲佐菌素建立糖尿病大鼠模型。中药1、2、3组大鼠分别给予各组中药灌胃,对照组及模型组给予等容积0.9%氯化钠注射液灌胃,每日1次,共6周。用酚红灌胃法检测胃排空率,免疫组织化学法观察胃组织胃促生长素的表达。结果与对照组比较,模型组、中药1、2、3组大鼠胃排空率、胃促生长素的表达明显降低(P<0.05);与模型组比较,中药1、2、3组大鼠胃排空率、胃促生长素的表达均明显升高(P<0.05);与中药3组比较,中药1、2组大鼠胃排空率、胃促生长素的表达均明显降低(P<0.05);中药1、2组间大鼠胃排空率、胃促生长素的表达无显著性差异(P>0.05)。结论活血化瘀通络中药能促进糖尿病胃轻瘫大鼠的胃排空及胃促生长素的表达,此可能为其治疗糖尿病胃轻瘫的机制。Objective To investigate the effect of traditional Chinese medicine for activating blood circula- tion and eliminating stasis on gastric emptying and ghrelin expression in rat with diabetic gastroparesis. Methods 50 healthy male SD rats were randomly divided into 5 groups; control group, model group, Chinese medicine group 1 （Salvia miltiorrhiza and Rhizoma Chuanxiong）, Chinese medicine group 2 （Leeches and Earthworm） and Chinese medicine group 3 （Salvia miltiorrhiza, Rhizoma Chuanxiong, Leeches and Earthworm）. Diabetes was induced by in- traperitoneal injection of streptozocin （STZ）, except for control group. And the latter three groups were treated with the Chinese traditional medicine which promote blood circulate and remove the stasis intragastrically once a day for six weeks. Gastric emptying was measured by phenol red and the expression of ghrelin was detected by immunohistochem- istry. Results Compared with control group, the gastric emptying and expression of ghrelin were decrease obviously in the other four groups（ P 〈 0.05 ）. Compared with model group, the gastric emptying and expression of ghrelin were in- crease obviously in each Chinese Medicine group （P 〈 0.05）. Compared with Chinese medicine group 3, the gastric emptying and expression of ghrelin decrease obviously in Chinese medicine groupl and 2 （ P 〈 O. 05 ）. There was no difference between Chinese medicine group 1 and Chinese medicine group 2 （ P 〉 0.05 ）. Conclusion The Chinese traditional medicine which promote blood circulate and remove the stasis can improve the gastric emptying and expres- sion of ghrelin, this might be its mechanism for the treatment of diabetic gastroparesis.
