欣通胶囊对麻醉犬血栓素B_2/6-酮-前列腺素及心肌梗死范围的影响

摘　　要：目的研究欣通胶囊对急性心肌梗死模型犬心肌的保护作用和机制,从而研究中药复方对冠心病的治疗方法,为临床开发新药提供依据。方法通过结扎犬冠状动脉左前降支复制急性心肌梗死模型,通过十二指肠给予受试药物,观察不同剂量组对模型犬血栓素、前列腺素、心脏定量组织学的影响。结果与假手术组比较,模型组TXB2/6-K-PG升高,4、6h差异有统计学意义(P<0.05,P<0.01)。与模型组比较,欣通胶囊中剂量组6h的TXB2/6-K-PG降低(P<0.05);欣通胶囊高剂量组、地奥心血康组在4、6hTXB2/6-K-PG降低,差异有统计学意义(P<0.05,P<0.01)。欣通胶囊高剂量组及地奥心血康组能够缩小心肌梗死范围。结论欣通胶囊对急性心肌梗死犬具有一定的保护作用,其机制可能是通过缩小心肌梗死面积,维持血栓素与前列腺素平衡等途径实现的。Objective To investigate the protective effect and mechanisms of Xintong capsule on the myocardial infarction in dogs, to study the traditional Chinese medicine for the treatment of coronary heart disease and provide a basis for the clinical development of new drugs. Methods The model of the myocardial infarction in dogs was developed by ligating the left coronary anterior descending artery and Administration of the test drug through the duodenum, observe the affect to the canine model in different dose groups thromboxaneB2, prostaglandins, heart quantitative histology and pathology. Results Model group compared with the sham group, the TXB2/6-K-PG increased, in 4 h and 6 h produce significant difference （P 〈 0.05, P 〈 0.01）. In 6 h TXB2/6-K-PG decreased by Xintong capsule middle dose group （P 〈 0.05）. TXBJ6-K-PG decreased in Xintong capsule high dose group and Di＇ao Xinxuekang group （P 〈 0.05, P 〈 0.01） in 4 h and 6 h. Xintong capsule high dose group and Di＇ao Xinxuekang group could narrow the scope of myocardial infarction. Conclusion Xintong capsule can significantly improve myocardial ischemia, reduce the myocardial infarction area. It possessed protective effect on myocardium. The mechanism may be realized through reduced myocardial infarct size, and a way to maintain thromboxaneB2 and prostaglandin balance.
