痰瘀同治方对肾间质纤维化大鼠肾组织CTGF表达的影响

摘　　要：目的:观察痰瘀同治方对肾间质纤维化大鼠肾组织结缔组织生长因子(CTGF)表达的影响,探讨其对肾间质纤维化的保护作用及作用机制。方法:实验采用UUO大鼠模型,将40只SD大鼠随机分为4组:假手术组、模型组、洛汀新治疗组、痰瘀同治方组。药物治疗14天后测定大鼠血清肌酐(SCr)、尿素氮(BUN)水平;肾组织HE、Masson染色观察病理改变;采用RT-PCR检测肾组织CTGFmRNA的表达;Western-blot检测肾组织CTGF蛋白表达。结果:与假手术组比较,模型组大鼠血清SCr、BUN水平显著升高,差异有统计学意义(P<0.01);肾组织CTGF表达显著升高(P<0.01,P<0.05),痰瘀同治方能够明显降低实验大鼠血清肌酐、尿素氮水平,能降低肾组织中CTGF表达,与模型组比较有显著性差异(P<0.01,P<0.05);形态学观察也显示其肾脏损害轻于模型组,效果优于洛汀新。结论:痰瘀同治方能通过下调CTGF的表达,延缓肾间质纤维化的进展。Objective: To observe the effect of prescription based on method of treating both phlegm and stasis at the same time on connective tissue growth factor(CTGF) expression in nephridial tissue of renal interstitial fibrosis rats,explore the protective effect on kidney and its mechanism. Methods: UUO models were applied,40 rats were divided into five groups randomly,sham operation group,model group,positive group(benazepril hydrochloride) ,xiaoyu xiezuoyin group and treating both phlegm and stasis group,each group was given the corresponding medicine for 14 days,than determining rats’SCr,BUN levels,taking nephridial tissue to make HE and Masson staining and observe the pathologic changes,determining CTGF mRNA expression of rats’nephridial tissue by RT-PCR,determining CTGF protein expression by Western Blot method. Results: To compare with sham group,Scr and BUN levels in model group were raised obviously,CTGF mRNA and protein expression increased significantly(P<0.01,P<0.05) ,prescription based on method of treating both phlegm and stasis and xiaoyu xiezuoyin could significantly lower SCr and BUN levels of rats with renal interstitial fibrosis,reduce CTGF mRNA and protein expression in rats’nephridial tissue,there were obvious difference between the treating both phlegm and stasis group and model group. The morphologic changes of rats’kidney in treating both phlegm and stasis group were significantly slighter than those in model group. Conclusion: Prescription based on method of treating both phlegm and stasis at the same time can delay the progression of renal interstitial fibrosis by down-regulating CTGF expression in nephridial tissue.
