摘 要:以27株镰孢菌菌株为试材,采用菌株形态学观察、rDNA-ITS序列分析以及寄主专化型鉴定相结合的方法,鉴定了尖孢镰孢菌番茄专化型。结果表明:经形态学和rDNA-ITS测序筛选,最后筛选出10株尖孢镰孢菌。将尖孢镰孢菌交叉接种到不同寄主植物,确定菌株编号YH070001和DL070009的镰孢菌为尖孢镰孢菌番茄专化型。与单一的传统方法相比,ITS rDNA方法有着较高的灵敏度,所以采取形态学、现代分子生物学以及传统交叉接种相结合的鉴定方式,能够使菌株专化型的鉴定更加准确、完善。Taking 27 Fusarium strains as materials,F.oxysporum f.sp.lycopersici was identified using morphology observation and rDNA-ITS sequencing.The results showed that after selected by morphology and rDNA-ITS sequencing,10 F.oxysporum strains were identified by traditional method of specialized form as cross inoculation.Thus N0.YH070001 and DL070009 were confirmed F.oxysporum f.sp.lycopersici.Compared with single traditional method,rDNA-ITS sequencing has higher sensitivity.To test the accuracy and feasibility of this identification method,morphology observation,rDNA-ITS sequencing and traditional method of specialized form identification were used in this study.
