摘  要:目的探讨负性情绪对卵清蛋白(OVA)致敏诱发哮喘大鼠细胞因子、NK细胞、神经激肽酶A受体(NK2-R)的影响及吸入糖皮质激素的干预效果。方法雄性sD大鼠32只分为4组,对照组(A组)、致敏组(B组)、恐惧应激致敏组(C组)、恐惧应激致敏+吸入糖皮质激素干预组(D组),给予OVA建立致敏大鼠模型;强迫浸水模拟恐惧应激环境干预;面罩雾化吸入糖皮质激素进行干预。采用四导生理记录仪测定各组大鼠呼吸道峰压值,HE染色观察肺组织呼吸道炎症,流式细胞仪测定外周血NK细胞数量,ELISA法测定血清IL-4、IFN-γ的水平,荧光定量PCR(RT-PCR)法测定大鼠肺组织中NK2-RmRNA水平。结果C组血清IL4水平及呼吸道峰压值高于B组及A组(P均〈0.05),而血清IFN-γ水平及外周血NK细胞数量较B组及A组降低(P均〈0.05);肺组织NK2-RmRNA水平c组较B组高(P〈0.05);D组呼吸道峰压值、血清IL-4水平及肺组织NK2一RmRNA水平均较C组降低(P均〈0.05),血清IFN-γ水平及外周血NK细胞数量D组较C组增高(P均〈0.05)。结论负性情绪能加重哮喘大鼠的呼吸道炎症,不仅影响Th1/Th2型细胞因子平衡,还能在减少外周血中NK细胞,增加肺组织NK2-RmRNA的表达等方面影响哮喘大鼠的呼吸道炎症;吸入糖皮质激素不仅能纠正Th1/Th2细胞因子失衡,降低肺组织NK2一RmRNA的表达,还能增加外周血NK细胞数量。Objective To determine the effect of negative moods on ovalbumin(OVA) in allergic rat models by examining cytokines and natural killer(NK) cells in peripheral blood, neurokinin A receptor(NK2-R) in the lung and observe the differentials during inhaled glucocorticoid inhalation. Methods Thirty-two healthy male SD rats were divided into 4 groups, group A as the control group, group B as the allergic group ,group C as the fear-stressed-allergic group, group D as the glucocorticoid inhalation treatment and fear-stress-allergic group. The allergic rat models were es- tablished by OVA. Forced swimming test was performed at the same time in group C and group D, and glucocorticoid in- halation was given at the same time as OVA challenge in group D. Airway peak pressure was measured with four-lead graph system, HE staining was used to observe the inflammation of the lung , the level of NK cells in peripheral blood was detected by flow cytometry, the cytokines IL-4, IFN-γ levels in the serum were determined by enzyme-linked immuno sorbent assay,NK2-R mRNA level was evaluated by transcription-quantitative. Results The levels of IL-4 and airway peak pressure in group C were higher than those in group B and group A ( all P 〈 0.05 ) ; the level of IFN-γ in the serum and the number of NK cells in peripheral blood in group C diminished compared with group B and group A ( P 〈 0.05) ; the expression of NK2-R mRNA in the lungs in group C was higher than that in group B(P 〈0.05) ; while in group D airway peak pressure, IL-4 level in the serum and the expression of NK2-R mRNA in the lungs were lower than those in group C( all P 〈0.05 ) ; the levels of IFN-γ in the serum and the number of NK cells in peripheral blood in group D were higher than those in group C ( all P 〈 0.05 ). Conclusions Negative moods may aggravate the inflammation reactions in the respiratory tract of the asthmatic rats, which may affect the balance between Th1/Th2 cytokines, reduce the number of NK cells in the periph
