摘 要:目的通过研究早期环境对发育期大鼠远期行为的影响,探讨下丘脑一垂体.肾上腺(HPA)轴在早期环境影响动物情绪、运动行为发育的作用。方法新生大鼠45只随机分为丰富环境(EE)组、隔离环境(IE)组及普通环境(NE)组,每组15只。EE组处于丰富化环境饲养,IE组大鼠为单调环境下成长,NE组大鼠常规饲养。大鼠于出生28d(P28)采用直线行走测试运动协调能力;于P29采用触动实验评价大鼠情绪行为,采用放射免疫学方法测定血清皮质酮。采用HE染色法检查脑神经元形态学。结果直线行走结果显示,大鼠运动协调能力:EE组〉NE组〉IE组(F=49.30,P〈0.000);触动实验结果显示,EE组大鼠紧张性刺激、触痛刺激评分(1.14±0.36,1.93±0.83)〈NE组(1.47±0.64,2.60±0.91)〈IE组(2.07±0.73,3.43±0.65)(P均〈0.01);基础血清皮质酮水平:EE组(32.56±9.05)肛g/L〉NE组(24.96±6.19)μg/L〉IE组(15.534-6.78)μg/L(F=19.71,P〈0.000);小剂量促皮质素(ACTH)激发试验后大鼠血清皮质酮水平:EE组(18.24±5.53)μg/L〈NE组(21.71±6.05)μg/L〈IE组(28.094-6.51)μg/L(F=10.25,P〈0.000)。HE染色结果显示,纹状体、杏仁核区神经元数量分别为:EE组[(72.80±6.81)个/HP,(73.53±6.14)个/HP]〉NE组[(65.67±8.98)个/HP,(61.47±6.57)个/HP]〉IE组[(53.33±6.84)个/HP,(48.13±6.53)个/HP](P均〈0.001)。结论早期环境可通过影响HPA轴功能而影响未成熟期大鼠情绪反应、运动行为及脑发育。隔离环境下成长的大鼠HPA轴功能失调,神经细胞受损,情绪、运动协调障碍;而早期丰富环境能缓冲大鼠在应激状态下HPA轴的过度活动,促进脑发育及改善远期行为。Objective To explore the effects of the function of hypothalamus-pituitary-adrenal (HPA) axis on emotional development and motor coordination ability by studying the influences of early environment on long-term be- haviors of developing rats. Methods Forty-five neonatal rats were randomly divided into 3 groups : the enriched envi- ronment group (EE group), the isolated environment group (IE group) and the normal environment group (NE group) ,with 15 eases in each group. There were 15 rats nurtured separately in EE group,IE group or NE group. The rat motor coordination abilities were tested by straight walking test on postnatal day 28 ( P28 ) and the emotional behaviors were evaluated by handling test at P29. The levels of serum corticosterone(CORT) of rats were detected by using emis- sion immunology method. The morphology of nenral cells in rat brain was examined by adopting HE staining. Results In the straight walking test, the motor coordination abilities of rats in EE group 〉 NE group 〉 IE group ( F = 49. 30, P 〈 0. 000). In the handing test,the scores of reaction for stressful stimuli and painful stimuli in IE group were ( 1.14±0.36,1.93±0.83) 〈 (1.47 ±0.64,2.60±0.91) inNEgroup〈(2.07±0.73,3.43±0.65) inlEgroup (allP〈 0.01 ). The base serum CORT levels were (32.56±9.05 ) Ixg/L in EE group 〉 ( 24.96±6.19 ) μg/L in NE group 〉 ( 15.53±6.78)μg/L in IE group ( F = 19.71, P 〈 0. 001 ). After a lower-dose adrenocorticotropic hormone stimula- tion test,the concentration of CORT was ( 18.24±5.53 )μg/L in EE group 〈 (21.71±6.05 )μg/L in NE group 〈 (28.09 ±6.51) Ixg/L in IE group (F = 10.25,P 〈0.1300), The HE staining showed the number of neural cells in striaturn and amygdale of rats were [ (72.80±6.81 ) cellfHP, ( 73.53±6.14 ) cell/HP ] in EE group 〉 [ ( 65.67 ±8.98) cell/HP,(61.47±6.57) cell/HP] in NE group 〉 [ (53.33 ±6.84) cell/HP, (48.13±6.53) cell/HP] in IE group �
