摘  要:目的:考察微粉化对制马钱子治疗类风湿性关节炎量效关系的影响。方法:以粗粉(超微粉碎入磨粒径)为对照,采用RA大鼠模型,以50.0,35.0,24.5,17.2,12.0mg·kg-1为给药剂量,每天给药一次,连续给药21 d,考察马钱子超微粉对模型大鼠的足趾容积、超氧化物歧化酶(SOD)、丙二醛(MDA)、一氧化氮(NO)、肿瘤坏死因子(TNF-α)的影响及量效关系。结果:马钱子超微粉与其粗粉均能抑制模型大鼠的足肿胀、升高SOD活性,降低MDA,NO,TNF-α含量,马钱子超微粉与其粗粉最小有效量分别为17.2 mg·kg-1、24.5 mg·kg-1;有效剂量范围分别是17.2~35.0mg·kg-1和24.5~50.0mg·kg-1。结论:微粉化后马钱子最小起效剂量降低、药理效应加强,临床使用宜降低剂量至1/2~2/3。OBJECTIVE To investigate the influence of micronization of Semen Strychni on the dose-effect relationship in rheumatoid arthritis rat model. METHODS The powder of Semen Strychni was made as the control, the rheumatoid arthritis (RA) rat model was used and administered once 50.0,35.0,24. 5,17. 2,12. 0 mg·kg^-1 dose per day for 21 d. The effects on swelling rate of hindpaw, superoxide dismutase (SOD), malondialdehyde (MDA), nitric oxide (NO), and tumor necrosis fac- tor (TNF-alpha) were observed, and the dose-effect relationship of ultrafine powder of Semen Strychni was investigated in the model rats. RESULTS The powder and ultrafine powder of Semen Strychni could inhibit the swelling of hindpaw, increased the activity of SOD, reduced the content of MDA, NO, TNF-α The minimum effective dose of ultrafine powder and powder of Semen Strychni was 17. 2,24. 5 mg·kg^-1 ; effective dose range was 17.2 to 35.0, 24. 5 to 50. 0 mg·kg^-1 , respectively. CON- CLUSION The minimum effective dose was reduced after micronization, and the pharmacological effect was increased, so we should reduce the dose to 1/2 -2/3 in clinic.
