High-throughput Screening of High Salinomycin-producing Strains with UV Treatment on Protoplasts

摘  要:以白色链霉菌(Streptomyces albus)S-610为出发菌株,利用溶菌酶水解菌丝体细胞壁获得原生质体,然后进行紫外诱变,并通过高通量筛选再生突变株。结果显示,当UV照射原生质体20 s时,细胞正突变率最大为24.5%,此时致死率为86.4%。经高通量筛选和摇瓶验证,最终筛得3株遗传稳定的高产菌S-612、S-620和S-622,产盐霉素能力较出发菌株S-610分别提高了57.6%、45.5%和69.7%。The protoplasts of Streptomyces albus S-610 were prepared by losozyme hydrolyzation and then were treated by UV. Regenerative mutants were screened by high-throughput screening procedure. Positive mutation rate of cells reached 24.5% of maximum value and lethality rate was 86.4% when the protoplasts were treated by UV for 20 s. Finally, 3 high producers with genetic stability, designated S-612, S-620 and S-22, respectively, were successfully obtained after high-throughput screening and validation in shake flasks. Compared with the original strain S-610, their salinomycin productions were increased by 57.6 %, 45.5 % and 69.7 %, respectively.
