摘  要:建立了五参二连颗粒的高效液相色谱指纹图谱及多指标成分(丹酚酸B、橙皮苷、哈巴俄苷和盐酸小檗碱)的含量测定方法。采用Agilent Zorbax SB-C18色谱柱,以乙腈-0.04 mol/L乙酸铵溶液(用冰乙酸调至pH 4.2)为流动相,梯度洗脱,检测波长270 nm。丹酚酸B、橙皮苷、哈巴俄苷和盐酸小檗碱分别在8~160、9.6~192、1.6~32和4.8~96 g/ml范围内线性关系良好,平均回收率分别为100.5%、99.8%、98.6%和100.1%,RSD分别为0.9%、1.2%、1.6%和1.1%。在指纹图谱研究中,以盐酸小檗碱为参照峰,标定了22个共有峰,10批五参二连颗粒的指纹图谱平均相似度大于0.96。An HPLC method was established for the determination of fingerprint of Wusen Erlian granules and the multi-component quantitative analysis of salvianolic acid B, hesperidin, harpagoside and berberine hydrochloride. An Agilent Zorbax SB-C18 analytical column was used with the mobile phase of acetonitrile-0.04 mol/L ammonium acetate solution (adjusted to pH 4.2 with glacial acetic acid) by gradient elution at the detection wavelength of 270 nm. The calibration curves of salvianolic acid B, hesperidin, harpagoside and berberine hydrochloride were linear in the concentration ranges of 8 - 160,9.6 - 192,1.6 - 32 and 4.8 - 96μg/ml, respectively. The average recoveries were 100.5 %, 99.8%, 98.6% and 100.1%, with RSDs of 0.9%, 1.2%, 1.6% and 1.1%, respectively. The fingerprints of Wusen Erlian granules were obtained, in which the peak of berberine hydrochloride was appointed as the benchmark peak. Twenty-two co-possessing peaks were selected as the fingerprint peaks. The fingerprint similarity values of the ten batches were above 0.96.
