摘 要:目的:研究尿毒清颗粒对糖尿病肾病(DN)模型大鼠肾组织的保护作用。方法:通过采用腹腔注射链脲佐菌素(60mg/kg)以复制大鼠DN模型。实验分为空白(等容生理盐水)、模型(等容生理盐水)与尿毒清颗粒(40mg/kg)组,灌胃给药,每天1次,连续8周。测定大鼠体质量、肾质量、肾质量/体质量指数、血糖、血尿素氮(BUN)、肌酐清除率(Ccr)和血β2-微球蛋白,并切片观察各组大鼠肾脏组织形态学的变化。结果:与空白组比较,模型组大鼠肾质量、肾质量/体质量指数、血糖、BUN、Ccr、血β2-微球蛋白水平显著升高(P<0.01或P<0.05),肾组织中肾小球基底膜增厚、系膜增生。与模型组比较,尿毒清颗粒组大鼠的肾质量、肾质量/体质量指数、血糖、BUN、Ccr、血β2-微球蛋白水平显著降低(P<0.01或P<0.05),肾组织病理学改变程度也有所减轻。结论:早期灌胃尿毒清颗粒对减轻肾小球肥大以及硬化程度具有一定的作用,从而可以延缓DN模型大鼠肾功能的衰减。OBJECTIVE: To study the protective effect of Niaoduqing granules on renal tissue in diabetic nephropathy (DN) rats. METHODS: DN model was established by intraperitoneal injection of streptozotocin (60 mg/kg). Model rats were divided in-to 3 groups, i.e. blank group (constant volume of normal saline), model group (constant volume of normal saline) and Niaoduqing granules group (40 mg/kg). They were given medicine intragastrically once a day for consecutive 8 weeks. The body weight, renal weight, renal weight/body weight index, blood glucose, BUN, Ccr and blood 152 microglobulin were determined. The morphological changes of renal tissue were observed with slice. RESULTS: Compared with blank group, renal weight, renal weight/body weight index, blood glucose, Bun, Ccr and blood 132 microglobulin were increased significantly in model group(P〈0.01 or P〈0.05). Patho-logical changes of renal tissue were observed, such as glomerular basement membrane thickening and mesangial cells proliferation. Compared with model group, renal weight, renal weight/body weight index, blood glucose, BUN, Ccr and blood 152 microglobulin of Niaoduqing granules group decreased significantly(P〈0.01 or P〈0.05) ; the pathological changes of kidney tissues were re-lieved to some extent. CONCLUSIONS: Niaoduqing granules may play a protective role against glomerular hypertrophy and sclero-sis so as to down-regulate renal function reduction of DN model rats.
