摘  要:目的观察益肾调气法对产后抑郁模型大鼠行为及脑海马区5-羟色胺(5-HT)、5-羟色胺1A受体(5-HT1AR)的影响。方法将雌性SD大鼠随机分为正常组、假手术组、模型组、颐脑解郁方组、氟西汀组5组,除正常组和假手术组外,其他3组采用激素刺激妊娠后停撤激素的方法建立产后抑郁大鼠模型。观察药物干预前后大鼠行为学的变化,并分别以高效液相色谱法和免疫组化法检测大鼠脑海马区5-HT的含量和5-HT1AR的表达变化。结果模型组在行为学测试中表现出糖水消耗减少、旷场实验得分减低、强迫游泳实验不动时间延长,有效模拟出抑郁样行为状态;而颐脑解郁方组、氟西汀组和模型组相比,上述行为状态改善明显。模型组双侧海马5-HT含量下降,海马CA3区5-HT1AR阳性细胞先增多后减少,颐脑解郁方组、氟西汀组和模型组相比,5-HT含量及5-HT1AR阳性细胞数均明显增多。结论益肾调气中药在产后抑郁大鼠模型上表现出明显的抗抑郁作用,其作用机制与调节脑组织海马区5-HT、5-HT1AR的表达有关。Objective To observe the influences of therapy of tonifying kidney and regulating qi on behaviors and levels of hippocampus 5-hydroxytryptamine (5-HT) and receptor of 5-HT 1 A ( 5-HT1A R) in rats with postpartum depression. Methods Female SD rats were randomly divided into normal group, sham-operation group, model group, Yinao Jieyu Fang group and fluoxetine group. Except of normal group and sham-operation group, other three groups were given hormone to stimulate pregnancy and then stopping the hormone for establishing rat model of postpartum depression. The changes of rat behaviors were observed before and after medicinal intervention. The changes in hippocampus 5-HT content and 5-HT1AR expression were detected by using high performance liquid chromatography (HPLC) and immunohistoehemistry technique. Results In model group, the behavior test showed that sucrose consumption decreased, scores of open - field test (OFT) were reduced and immobility time of forced swimming test (FST) was prolonged, so a depressive behavior state was effectively simulated. All of the above-mentioned changes of behaviors were relieved in Yinao Jieyu Fang group and fluoxetine group compared with model group. The 5-HT content in bilateral hippocampus decreased and positive cells of 5-HT1AR in hippocampus CA3 zone increased at first and then decreased in model group. The 5-HT content and positive cells of 5-HTIA R increased significantly in Yinao Jieyu Fang group and fluoxetine group compared with model group. Conclusion The TCM therapy of tonifying kidney and regulating qi had significant anti-depression effect in rat model of postpartum depression, and the mechanism may be related to the regulation of hippoeampus 5-HT content and 5-HT1AR expression.
