摘  要:为延长硫酸链霉素的作用时间,提高其对烟草青枯病的抗病能力并减少其残留,利用壳聚糖与γ-聚谷氨酸间的静电自组装作用形成纳米微胶囊,在成囊过程中将硫酸链霉素负载其中,制备了壳聚糖/γ-聚谷氨酸负载硫酸链霉素纳米微胶囊,并对不同硫酸链霉素用量及壳聚糖和γ-聚谷氨酸的质量比对成囊效果及负载效果的影响进行了研究。结果表明:随着硫酸链霉素添加量的增加,微胶囊的载药量增大;随着壳聚糖与γ-聚谷氨酸质量比的减小,不同硫酸链霉素添加量下,包封率均呈增大趋势,同时载药量均呈降低趋势;当硫酸链霉素添加量为10 mg,m(壳聚糖)∶m(γ-聚谷氨酸)=20∶17时,包封率最高(85.2%),载药量达46.8%。傅立叶变换红外光谱分析及差示扫描量热法分析结果表明,微胶囊对硫酸链霉素负载效果较好;透射电子显微镜下,载药纳米微胶囊呈规整实心球形,粒径分布较均匀,平均粒径在20~50 nm之间;模拟释放结果表明,纳米微胶囊对硫酸链霉素具有缓释效果。To prolong streptomycin sulfate effective time, streptomycin sulphate-loaded nanocapsule coated with chitosan and γ-polyglutamic acid was prepared and characterized. The composite nanocapsule was prepared using a simple, mild ionic-gelation method, and streptomycin sulphate was loaded during gelating. The effects of streptomycin sulfate dosage and chitosan/poly-γ-glutamic acid mass ratio on the process of encystation were tested. The results indicated that loading efficiency increased with increasing of streptomycin sulfate dosage, encapsulation efficiency increased and loading efficiency decreased when the mass ratio of chitosan and γ-polyglutamic acid decreased. When the dosage of streptomycin sulfate was 10 mg, and the mass ratio of chitosan to γ-polyglutamic acid was 20: 17, the highest encapsulation efficiency (85.2%) was gotten, and loading efficiency reached 46. 8%. The Fourier transform infrared spectroscopy and differential scanning calorimetric analysis confirmed that streptomycin sulfate was loaded in nanocapsule, and the solid nanocapsule with average diameter 20-50 nm was observed under transmitted electronic microscopy. Sustained-releasing of streptomycin sulfate was observed under simulation releasing test.
