摘 要:目的探讨山楂叶总黄酮对慢性脑缺血大鼠学习记忆及肿瘤坏死因子-α(TNF-α)、白介素-1(IL-1)含量的影响。方法成年雄性SD大鼠32只,随机分为假手术组、模型组、山楂叶总黄酮组和尼莫地平组,每组8只。采用永久性双侧颈总动脉结扎法建立慢性脑缺血模型,山楂叶总黄酮140 mg/(kg.d),连续灌胃给药36 d。采用八臂迷宫评价大鼠学习记忆能力,放射免疫法检测脑组织TNF-α、IL-1水平含量。结果与假手术组比较,模型组大鼠出现明显的学习记忆障碍,学习记忆和工作记忆错误系数显著增多(P<0.01),脑组织TNF-α、IL-1含量显著增加(P<0.01);与模型组比较,山楂叶总黄酮能改善慢性脑缺血大鼠的学习记忆障碍,减少大鼠学习记忆和工作记忆错误次数(P<0.05),降低脑组织TNF-α、IL-1含量(P<0.05);与尼莫地平组比较,山楂叶总黄酮组大鼠学习记忆和工作记忆错次数减少,脑组织TNF-α、IL-1含量降低,但无统计学差异(P>0.05)。结论山楂叶总黄酮通过降低TNF-α、IL-1含量,减轻脑组织炎症反应,从而提高慢性脑缺血大鼠学习记忆能力。Objective To explore the efficacy of total flavanone of hawthorn leaf in treatment of memory impairment and inflammatory reaction in rats with chronic cerebral ischemia.Methods Sixty four SD male rats were randomly divided into sham operation group,model group,total flavanone of hawthorn leaf group and nimodipine group.Each group had 16 rats.The model of chronic cerebral ischemia was established by permanent ligation of bilateral carotid arteries.Total amount of flavanone of hawthorn leaf at dosage of 140mg / kg.d per rat was intragastrically administered for 36 days.The ability of learning and memory in these rats was evaluated by the eight-arm maze method.The content of TNF α and IL-1 in brain tissue was measured by radioimmunoassay.The expression of GFAP in brain tissue was detected by immunohistochemical method.Results The application of total flavanone of hawthorn leaf can improve the impairment of learning and memory in rats with chronic cerebral ischemia and reduce the number of errors in learning memory and working memory of rats(P 0.05),inhibit GFAP expression in brain tissue(P 0.05),and reduce the content of TNF-α and IL-1 in brain tissue(P 0.05).Conclusion Total flavanone of hawthorn leaf can reduce the content of TNF-α and IL-1 and decrease the inflammatory response of brain tissue by inhibition of GFAP expression in rats with chronic cerebral ischemia,thus,it may improve the capability of learning and memory in rats with chronic cerebral ischemia.
