摘  要:目的:评价他克莫司联合罗格列酮对TNF-α诱导的HaCaT细胞体外增殖和分化的影响。方法:他克莫司(10/maol/L)和不同浓度罗格列酮单独及联合作用于TNF-α诱导的HaCaT细胞48h后,用CCK-8法测定细胞增殖,逆转录一聚合酶链反应(RT—PCR)检测增殖分化标志物角蛋白6及外皮蛋白mRNA的表达水平;结果:他克莫司和罗格列酮均可抑制TNF-α诱导的HaCaT细胞体外增殖,下调角蛋白6mRNA表达,上调外皮蛋白mRNA表达,且联合用药组的作用更强。各组细胞生长抑制率:他克莫司10μmaol/L组为(21.71±5.36)%,罗格列酮组10、20、40、80μmol/L组分别为(5.04±7.12)%、(12.56±6.93)%、(21.41±5.95)%和(39.17±8.03)%,联合1-4组分别为(27.60±6.68)%、(38.35±7.24)%、(48.60±8.40)%和(65.21±4.53)%。结论:他克莫司、罗格列酮单独或联合应用均可抑制TNF-α诱导的HaCaT细胞体外增殖,促进其分化。Objective: To determine the effect of combined tacrolimus with rosiglitazone on the proliferation and differentiation of TNF- ainduced HaCat cells. Method: After TNF- alpha - induced HaCaT cells were treated with tacrolimus ( 10 μmol/L) and rosiglitazone ( 10, 40 μmol/L) single or in a combined way for 48 hours, the prolifera- tion of the cells were detected by CCK- 8 assay, and the expression of kerain6 and involucrin mRNA were semi - quantitatively analyzed by reverse transcription polymerase chain reaction (RT- PCR). Results: After 48 hours, the proliferation inhibition rate of combined group was significantly higher than single groups. Involucrin mRNA expres- sion was significantly up- regulated, keratin6 mRNA expression significantly down- regulated and the combined groups had stronger effect on the expression. Conclusion: Tacrolimns and rosiglitazone can inhibit the proliferation of TNF- alpha- induced HaCaT cells and induce the differentiation in single or combined way.
