摘  要:目的:评价积雪草苷对培养的原代瘢痕组织成纤维细胞(fibroblast,FB)的细胞增殖活性、自分泌Ⅰ、Ⅲ胶原和转化生长因子p1(transforming growth factorp1,TGF-β1)mRNA的影响。方法:体外培养原代FB,加入不同浓度(0.01、0.05、0.1、0.5、1.0mg/mL)积雪草苷,观察瘢痕FB的形态变化;并用M3T评估不同浓度积雪草苷对瘢痕阳增殖的抑制作用;运用RT-PCR检测Ⅰ、Ⅲ型胶原和TGF-β1 mRNA的表达。结果:与对照组相比,积雪草苷浓度在0.1-1.0mg/mL时,瘢痕FB体积缩小、变短,树突状突起回缩,细胞间隙变大,并呈浓度依赖模式。结论:一定药物浓度范围内,积雪草苷可干预瘢痕形成。Objective: To determine the effects of asiaticoside on the expression of autocrine Ⅰ,Ⅲ collagen and transforming growth factor -β1 (TGF-β1 ) mRNA in cultured keloid fibrohlast cell ( KFB ). Method: After FB was cultured and asiaticoside added in different concentrations (0.01, 0.05, 0.1,0.5 and 1.0 mg/mL), the morpho- logical changes of FB observed. The inhibitive effect of asiaticoside on the FB proliferation was measured by MTT as- say. The expression of type Ⅰ, and Ⅲ collagen and TGF -β1 mRNA was detected by RT - PCR. Results: Com- pared with control group, FB volume was reduced, shortened, dendritic neurite was retracted and the cells gap be- came larger when the concentration of asiaticoside in 0.1-1 mg/mL added. These effects were in a concentration dependent manner. Conclusion: Within certain concentration, asiaticoside can intervene the formation of keloid.
