摘 要:用小鼠Langendorff心脏灌流法 ,测定保存前后的心功能指标及心肌含水量。动物随机分成两组 ,对照组使用mEC保存液 ,粉防己碱组 (TET组 )使用含粉防己碱 0 .0 3mmol/L的mEC液。保存时间为 4h。结果表明 :TET组保存后再灌注 30min、4 0min、6 0min时LVPSP、+DP/DTmax优于mEC组 ,差异显著 (P <0 .0 5) ,TET组保存后再灌注 30min、6 0min时 ,-DP/DTmax显著优于mEC组 (P <0 .0 5)。保存后再灌注 30min、4 0min、6 0min时 ,TET组RPP优于mEC组 ,差异显著(P <0 .0 5)。表明粉防己碱可以明显提高mEC液的心脏保存效果 ,TET组也可以提高保存后心脏的心率和冠脉流量 。Objective: To observe the effects of Tetrandrine(TET) on heart preservation. Methods: Hearts from Kunming mice were perfused on a Langendorff apparatus. After 5 minutes' stabilization, baseline measurements of heart rate, coronary flow, LVPSP, DP/DT were performed. Hearts (n=7 in each group) were then randomized to two groups, TET group with 0.03 mmol/L TET added into modified Euro Collins (mEC)solution, and mEC group. After perfusion, hearts were stored in preservation solution for 4 hrs at 4℃, then remounted on the perfusion apparatus, and stabilized as before, cardiac functions were then repeated. Results: Compared with mEC group, TET group has significant effects on systolic and diastolic function of heart. Conclusion: The addition of TET to mEC solution is associated with significantly enhanced cardiac function in the isolated mouse heart after 4 hrs of simple cold storage, but TET group cannot improve the heart rate and coronary flow. These data suggest the usage of the extracts of Chinese Medicinal herbs in heart preservation.
