摘 要:建立川楝子水提醇沉提取物中川楝素含量测定的反相高效液相色谱法。采用Diamonsil TMC18柱(250×4.6 mm,5μm);流动相为乙腈-水(40∶60);流速为1.2 mL.min-1;检测波长为220 nm,柱温为25℃,进样量20μL。川楝素检测浓度在(10.39~249.36)μg.mL-1范围内与川楝素峰面积呈良好的线性关系(r=0.999 9,n=6);平均加样回收率为99.52%,RSD为0.81%。建立了简便、快速、准确、重复性好的川楝子水体醇沉提取物中川楝素的RP-HPLC含量测定方法。An RP-HPLC method is established for the determination of toosendanin in water extraction and eth- anol precipitate (WEP) of the Melia toosendan Sieb. The RP-HPLC system consisting of Diamonsil T^M C18 (250 × 4.6 mm,5μm) column was adopted and the mobile phase was acetonitrile-water (40: 60) at the flow rate of 1.2 mL · min- 1 ; The detection wavelength is 220 nm and the column temperature is 25 ℃ and injection volum is 20μL, using external standard method. The method has a good linearing in the ranges in the ranges of 10. 39 249.36μg · mL^-1 (r =0. 999 9 ,n =6) for the sum of two signal peaks of toosendanin. The intra-day RSDs of high, medium and low concentration were 0. 63 %, 0. 54 %, 0. 90 %, the inter-day RSDs were 1.61%, 1.40 %, 1.40 %. The average recovery was 99.52 % (RSD =0. 81% ). The RP-HPLC method proposed for the determi- nation of toosendanin in Melia toosendan Sieb extracted by water and precipitated by ethanol is simple, rapid, ac- curate, reproducible, and can be applied for the determination of samples.
