摘  要:目的研究促血小板生成素(thrombopoietin,TPO)的心肌保护作用。方法小鼠分为对照组、多柔比星组、TPO组。多柔比星组及TPO组小鼠第1天予多柔比星20 mg/kg经腹腔注射;TPO组小鼠予TPO 15μg/kg,共3次经腹腔注射。第1天(多柔比星注射前)和第5天予小鼠做超声心动图检查;第5天取小鼠心肌组织行苏木素伊红染色,显微镜下评价心肌组织损伤程度。比较第1~8天内多柔比星组和TPO组存活率。结果与第0天相比,第5天多柔比星组的心率、每分钟心输出量、左心室短轴缩短率明显下降,差异有统计学意义(P<0.05);镜下见小鼠心肌组织明显损伤,说明药物性心肌损伤的模型能成功建立。相对多柔比星组,TPO组心率、每分钟心输出量、左心室短轴缩短率明显改善,差异有统计学意义(P<0.05)。TPO组镜下评分示心肌损伤显著降低,8 d内的生存率明显高于多柔比星组,差异有统计学意义(P<0.05)。结论 TPO对损伤心肌有保护作用。Objectives To investigate the protective effect of thrombopoietin (TPO) on myocardium. Methods Cardiac dysfunction in mice model was induced by a single intraperitoneal injection of doxorubiein (20 mg/kg) in TPO group and doxorubicin group on the first day. TPO was injected in TPO group at a dose of 15 Ixg/kg for 3 times. Heart rate and heart function were detected by echocardiography at baseline and the fifth day. Pathological grade of cardiac tissue morphology was scored according to myofibrillar loss and cytoplasmic vacuolization by doxorubicin eosin staining. Survival rate in 8 days between the two groups was observed. Results Heart rate (HR), cardiac output (CO) and left ventricular fraction shortening (FS) sharply reduced (P〈0.05) and myocardium tissue was dramatically injured at the fifth day in doxorubicin group when compared with those of baseline, which indicated that the myocardium injured mise model was successfully built. Compared with doxorubicin group, HR, CO and FS obviously improved (P〈0.05), pathological grade of cardiac tissue morphology significantly decreased, survival ratio in 8 days was significantly higher in TPO group (P〈0.05). Conclusions TPO has protective effect on injured myocardium.
