摘 要:建立整体化色谱柱分离分析甘草酸18H-差向异构体的方法。采用C18整体化色谱柱,流动相为磷酸盐缓冲液-乙腈体系,探讨了磷酸盐缓冲液pH及浓度、乙腈含量、柱温等对甘草酸18H-差向异构体分离的影响,在优化色谱条件下,18α-、18β-甘草酸得到基线分离,分离度可达2.75,二异构体出峰时间在10 min内,且在2.12~21.2μg/mL范围内线性关系良好。运用整体化色谱柱缩短了甘草酸18H-差向异构体的分析时间,提高了分析效率,方法可运用于评价甘草酸的异构化及质量控制。A HPLC separation method for the analysis of the epimers of glycyrrhizic acid with a monolithic column was es- tablished. A chromolith RP-18α column was used,the mobile phase was phosphate buffer solution-acetonitrile system. The effects of pH value, phosphate buffer concentration, acetoni- trile content and column temperature on the separation of the epimers were investigated. Under the optimized condition, baseline separation for the two epimers was achieved. The res- olution was 2. 75, and the runtime was 10 minutes. The cali- bration curves were linear in the range of 2. 12 ~ 21.2 μg/ mL. The analysis efficiency was improved with this monolithic column,and this method can be used to track the isomeriza- tion reaction of glycyrrhizic acid.
