摘 要:微孢子虫营专性细胞内寄生,其分泌蛋白是一类重要的侵染互作因子。利用分泌蛋白预测程序EuSecPred 2.0对家蚕微孢子虫、中蜂微孢子虫、兔脑炎微孢子虫和毕氏肠道微孢子虫的分泌蛋白进行全基因组预测,分别获得了315、90、30和45个分泌蛋白。不同微孢子虫分泌蛋白的序列平均长度和氨基酸组成特征相似,信号肽和非信号肽区均以疏水性氨基酸为主;但统计所获蛋白质功能注释的分布却是大部分分泌蛋白为各种微孢子虫的特异蛋白,仅有少部分分泌蛋白为3种以上微孢子虫所共有,如几丁质脱乙酰基酶(CDA)、热激蛋白70(HSP70)、孢壁蛋白(SWP)和Ricin B-凝集素等。4种微孢子虫分泌蛋白信号肽区的基序分别为[LI]xx[IV]xAS、L[FY]x、[LI]LL[LI][GSA]L[VI][IS][CAG]和L[LF]LFLAISAGA[SA],非信号肽区域的基序分别为[LM][RK]N、L[RK][ND]、[KR][KR][KR]LP和[PG]D[LM]。研究结果有助于揭示微孢子虫的适应性进化机制,同时也为微孢子虫的侵染和与宿主互作机制的研究提供了靶标。Microsporidia are specialized intracellular parasites.Secretory proteins of microsporidia are important factors related to the infection and interaction with host cells.In this study,we predicted the secretory proteins of Nosema bombycis,Nosema ceranae,Encephalitozoon cuniculi and Enterocytozoon bieneusi at whole genome level using secretory protein prediction program EuSecPred 2.0 and obtained 315,90,30 and 45 candidates,respectively.Most secretory proteins from these four microsporidian species share similar features on average length and amino acid composition,and the signal peptide and non-signal peptide regions are mainly composed of hydrophobic amino acids.However,most secreted proteins of each microsporidian species are species-specific proteins based on their functional annotation by statistical analysis.Only a few secretory proteins are shared by three or more microsporidian species,e.g.,chitin deacetylase(CDA),heat shock protein 70(HSP70),spore wall protein(SWP) and ricin B-lectin.The motifs in signal peptide of secretory proteins from four microsporidian species was xxxAS,Lx,LLL,and LLFLAISAGA,respectively.The motifs in non-signal peptide of secretory proteins from four microsporidian species was N,L,LP,andD,respectively.These results will contribute to exploration of the mechanism of microsporidian adaptive evolution and provide targets for research on microsporidian infection and interaction with hosts.
