摘 要:目的:采用响应面法优化土鳖虫蛋白质的酶解工艺条件。方法:采用甲醛电位滴定法测定氨基氮的含量。以土鳖虫蛋白水解产物氨基氮的含量为指标,考察土鳖虫酶解的最适酶种(胃蛋白酶、胰蛋白酶和木瓜蛋白酶),并在单因素试验的基础上,采用Box-Benhnken响应面分析法,考察酶解温度、pH值和固液比等3个因素对氨基氮含量的影响,得到描述土鳖虫蛋白水解产物氨基氮含量与操作参数之间的2次响应面模型,确定土鳖虫蛋白质的最优酶解工艺条件。结果:最佳工艺条件为:选择胰蛋白酶为最适酶种,在固液比为1∶7、PH为5.0、酶解温度为60℃的条件下,酶解1 h。经验证试验此条件下氨基氮的得率为1.22%,与理论值(1.25%)基本相符。结论:该工艺成本低,蛋白质水解彻底,获得的氨基氮含量高,工艺稳定,重现性好,可为土鳖虫蛋白水解的工业化生产提供科学依据。Objective : To optimize the hydrolysis conditions of Eupolyphaga protein by using response surface methodology. Methods: Formaldehyde potentiometric titration was used to select the optimal enzyme from Pepsin,Trypsin and Papain. Amino nitrogen content was chosen as the indicator of Eupolyphaga protein hydrolyzate. Based on the single - factor test, Box - Benhnken response surface method was established for analysing influence factors of amino nitrogen, including hydrolysis temperature, pH and solid - liquid ratio. The quadratic response surface model between amino nitrogen content and operating parameters was gained and the optimal hydrolysis conditions of Eupolyphaga protein was confirmed. Results : The optimal hydrolysis conditions were as follows : the ratio of material to liquid was 1 : 7, pH was 5.0, hydrolysis tem- perature was set at 60℃ and enzymatictime was lh. The amino nitrogen yeild was 1.22% in this condition, which was consistent with the theoretical value of 1.25 %. Conclusion: The optimal hydrolysis condition has the advantage of low expense, full protein hydrolysis, obtaining high content of amino nitrogen and better stability and reproducibility. Therefore, this paper could provide a scientific basis on the industrial production of Eupolyphaga proteolytic.
