摘 要:目的:探讨电针"内关"穴对心肌肥厚模型大鼠一氧化氮(nitric oxide,NO)的影响。方法:SD大鼠50只随机数字表法分为正常组、模型组、电针组、p38抑制剂组、电针+p38抑制剂组,采用皮下注射盐酸异丙肾上腺素3 mg/(kg.d)建立心肌肥厚大鼠模型,电针组采用连续波,频率2 Hz,强度1 mA,通电20 min,1次/d,p38抑制剂组在造模基础上0.3 mg/(kg.d)SB203580皮下注射,共14 d,电针+p38抑制剂组在造模基础上同时应用抑制剂和电针,大鼠于第15 d测量左心室壁平均厚度、心肌细胞平均直径、心肌纤维横径,硝酸还原酶法测定血清和心肌组织NO含量,并对数据进行统计分析。结果:模型组大鼠左心室壁平均厚度、心肌细胞平均直径、心肌纤维横径于造模后升高,心肌组织和血清NO含量造模后降低,和正常组比较,有非常显著性差异(P<0.01);经电针和抑制剂治疗后大鼠左心室壁平均厚度、心肌细胞平均直径、心肌纤维横径均明显低于模型组,NO均明显高于模型组,和模型组比较有显著性差异(P<0.05,P<0.01),3组之间比较无显著性差异(P>0.05)。结论:电针"内关"可以明显抑制心肌肥厚,其抗心肌肥厚作用可能是通过升高NO来实现的。Objective:To discuss the influence of electroacupuncture at Neignan on NO in rats with cardiac hypertrophy. Methods : Fifty SD rats were randomly divided into five groups by random number table method,with ten rats in each group respectively. Rats in model group were received 3 mg/(kg . d)of isoprenaline hydrochloride. The electroacupuncture group selected a stimulus parameter: continuous - wave, frequency of 2 Hz, current intensity of 1 mA, and treated for twenty minutes, once a day. The P38MAPK inhibitor group were injected 3 mg/( kg . d) of isoprenaline hydrochloride and 0.3 mg/(kg . d)of SB203580. The electroacupuncture and P38MAPK inhibitor group were treated by both electroacu- puncture and P38MAPK inhibitor, amounted to 14 days. The average measurement of left ventricular wall thickness, the average diameter of the myocardial cell, and the myocardial fiber diameter were measured. The contents of NO in heart tissue and blood plasma were determined by the method of nitric acid reductase, then the data was analyzed through an statistical analysis system. Results : Compared with the normal group, the average measurement of left ventricular wall thickness, the average diameter of the myocardial cell and the myocardial fiber diameter were significantly higher with statistical significance in model group(P 〈 0.01 ), the contents of NO in heart tissue and blood plasma were significantly lower with statistical significance in model group (P 〈 0.01 ). After the treatment, the average measurement of left ventricular wall thickness,the average diameter of the myocardial cell and the myocardial fiber diameter were significantly lower and NO levels in heart tissue and blood plasma were significantly higher with statistical significance in the electroacupuncture group, the P38MAPK inhibitor group, and the electroacupuncture and P38MAPK inhibitor group ( P 〈 0.05, P 〈 0.01 ), but there was no significant difference between them( P 〉 0.05).
