摘 要:目的:观察丹参多酚酸盐对肝硬化大鼠肝组织TNF-α和IL-6 mRNA表达影响。方法:SD大鼠40%CCL4-橄榄油溶液建立肝硬化大鼠模型,随机分成4组:模型组、丹参多酚酸盐治疗低、中、高剂量组,分别每天腹腔注射葡萄糖溶液、丹参多酚酸盐12、24、48 mg/kg 2周。另取10只大鼠正常对照。治疗结束后观察各组大鼠死亡率,肝组织外观,光度法测门静脉血内毒素水平,光镜下观察病理形态学改变,Real-time RT-PCR法检测肝脏组织TNF-α和IL-6 mRNA的表达变化。结果:模型组大鼠死亡率42.86%,丹参多酚酸盐高剂量组无死亡率;与正常组比较,模型组内毒素水平显著升高(P<0.01),经丹参多酚酸盐治疗后低剂量组内毒素水平降低(P<0.05),丹参多酚酸盐中剂量组与高剂量组显著降低(P<0.01)。丹参多酚酸盐治疗组肝脏色泽、质地、边缘、表面光滑度及组织病理较模型对照组有明显改善。高剂量组TNF-αmRNA和各治疗组IL-6mRNA表达量与模型组相比有显著统计学差异(P<0.01),中剂量组TNF-αmRNA表达量与模型组相比有统计学差异(P<0.05)。结论:丹参多酚酸盐降低肝硬化大鼠死亡率,改善肝硬化大鼠肝组织炎症程度,抑制肝组织TNF-α和IL-6 mRNA表达。Objective : To investigate effect of salvianolate on cytokine gene expression in liver tissue of rats with CCl4 - induced liver cirrhosis. Methods:Cirrhosis was induced in rats using CCl4 in a 2:3 mixture with olive oil (0.3 mL/kg). Rats were divided randomly into four groups:untreated,low -dose salvianolate (12 mg/kg), medium -dose salvianolate (24 mg/kg) ,and high -dose salvianolate(48 mg/kg)groups. They were all treated for 2 wk. Another 10 healthy rats were enrolled as normal controls. Mortality of cirrhotic rats in each group was evaluated after treatment. Serum samples were collected from the portal vein for the detection of endotoxin. Liver tissue samples were observed under light microscopy for morphological changes. Expressions of TNF - a and IL - 6 mRNA in the liver tissue were analyzed by Real - time reverse - transcriptase polymerase chain reaction. Results : The mortality of cirrhotic rats in the untreated group was 42.86%. No cirrhotic rat died in the high - dose salvianolate group. Serum endotoxin level was higher in the untreated group than that in salvianolate - treated group. Salvianolate therapy induced improvement in liver histopathological changes. The TNF-a and IL- 6 mRNA expression levels in liver tissue were significantly lower in different salvianolate - treated groups than in the non - treatment group. Conclusion : Salvianolate can reduce endotoxin level, improve liver histopathological changes, and inhibit the expressions of TNF - a and IL - 6 mRNA of liver tissue in cirrhotic rats.
