摘  要:目的:通过研究活血化瘀中药对TGF-β1/Smad3通路的影响,探讨活血化瘀法对急性心梗后心肌重塑的效应机制。方法清洁级Wistar大鼠50只随机分为5组,分别为正常组、模型组、假手术组、活血化瘀组、AMD3100组,每组10只。祛瘀生组大鼠术前3 d给药,1次/d。7 d后,实时荧光定量RT-PCR检测心肌组织转化生长因子(TGF)β1mRNA、smad3mRNA表达,分析各组非梗死区心肌胶原含量,并观察心肌组织H-E染色结果。结果:模型组心肌组织TGF-β1mRNA、smad3mRNA表达均增加(P<0.05);活血化瘀组能降低心肌组织TGF-β1mRNA、smad3mRNA表达水平,降低心肌胶原含量,并能减轻心肌组织的病理损伤;活血化瘀组与AMD3100组相比较TGF-β1mRNA、smad3mRNA表达无明显差异(P>0.05)。结论:活血化瘀法可能通过对TGF-β1/Smad3通路的抑制作用,从而有效的抑制急性心梗后心肌重塑。Through the study of the relationship between TGF-βl/Smads and ventricular remodeling after a- cute myocardial infarction in rats treated with Eliminating Blood Stasis to Promote Regeneration of Blood, investigate the Ef- fects of Eliminating Blood Stasis to Promote Regeneration of Blood in Ventricular Remodeling after AMI, and provide experi- mental basis. Methods: 50 rats were randomly divided into five groups: Myocardial infarction group, sham - operated group, Eliminating Blood Stasis to Promote Regeneration of Blood treatment group, blank control group, AMD3100 group. Each group has ten rats. Eliminating Blood Stasis to Promote Regeneration of Blood treatment group were administered 3 days be- fore the operation. The rats were sacrificed 7 days later. To observe the mRNA level of transforming growth factor (TGF)β1, Smad3 and Smad7 with RT-PCR, non infarcted myocardial collagen content and H-E staining results in colon. AMD3100 group and Eliminating Blood Stasis to Promote Regeneration of Blood treatment group were same( P 〉 0.05).Results: The expressions of TGF-β1 mRNA and Smad3 mRNA were higher in model - group than sham group( P 〈 0.05) and the expres- sion of Smad7 mRNA in these areas was decreased in AMI group which compared with sham group. Eliminating Blood Stasis to Promote Regeneration of Blood could reduce the expression of TGF-β1 mRNA and Smad3mRNA and myocardial injury, non infarcted myocardial collagen content and increase Smad7mRNA. Conclusion: Eliminating Blood Stasis to Promote Re- generation of Blood has inhibitory effect in ventricular remodeling after myocardial infarction.
