摘 要:目的:探讨miR-155表达水平在链脲佐菌素(STZ)诱导糖尿病肾病(DN)大鼠及大鼠肾系膜细胞中是否存在同样的变化。方法:将雄性清洁SD大鼠8只随机分成实验组(DN组,n=4)和对照组(NC组,n=4),DN组大鼠腹腔注射足量STZ诱导DN大鼠,观察血糖及肾脏结构变化,用real-time PCR检测肾脏miR-155表达水平;大鼠肾系膜细胞分为高糖培养组和正糖培养组,培养24 h、48 h、72 h,用real-time PCR检测miR-155表达水平。结果:与对照组相比,DN组大鼠肾脏miR-155表达水平下降(P<0.05);高糖培养的大鼠肾系膜细胞miR-155表达水平较正糖培养的表达水平下降(P<0.05),于48 h表达水平最低。结论:miR-155在高糖培养大鼠肾系膜细胞表达下降。Objective: To explore the miR-155 expression of kidney from strepozotocin (STZ) induced rat with diabetic nephropathy (DN) and cultured rat mesangial cells. Methods: 8 male SD rats were randomly divided into control group and experimental group (4 for each). 4 experimental rats were injected with STZ into abdominal cavity to induce DN modal. Detect serum glucose, the renal pathology and miR-155 expression of kidney by real-time PCR. And cultured rat mesangial cells (RMCs) were maintained with normal glucose (NG) or exposed to high glucose (HG) for 24 hs, 48 hs and 72 hs, then the cells were harvested for real-time PCR to examine the expression of miR-155. Results: Compared with the control group, the miR-155 expression of kidney from rat with DN decreased significantly in the experiment group (P〈0.05). The expression of miR-155 in cultured rat mesangial cells exposed to high glucose also reduced compared to the cells maintained in normal glucose (P〈0.05). Conclusion: miR-155 decreased cultured rat mesangial cells exposed to high glucose.
