摘 要:目的:探讨丹参、地西泮联合应用对小鼠的抗焦虑作用及其作用机制。方法:将雄性小鼠随机分成7组,分别为模型组,地西泮高、中、低剂量组,丹参地西泮高、中、低剂量组。模型组灌胃0.5%CMC-Na,其他组灌胃相应药物,连续给药5d。末次给药0.5h后,记录小鼠进入开臂的次数占总入臂次数的百分比(OE%)及在开臂中滞留的时间占总入臂时间的百分比(OT%)。明暗箱实验记录小鼠10min内在明暗箱中的穿箱次数。观察丹参、地西泮联合应用对小鼠焦虑的干预作用。用酶免法测定小鼠脑内γ-氨基丁酸,谷氨酸,5-羟色胺的含量。结果:丹参地西泮高剂量组能显著增加小鼠进入高架十字迷宫OE%、OT%,显示出对小鼠的抗焦虑作用。丹参地西泮中剂量组能显著增加小鼠进入高架十字迷宫OT%和小鼠明暗穿箱次数,二者联用有协同增效的趋势。结论:丹参、地西泮联合使用有增加抗焦虑作用的趋势,其抗焦虑机制可能与能降低小鼠脑内的GLu含量有关。Objective:To explore the anti-anxiety effect and its mechanism of the combined application of salvia miltiorrhiza and diazepam on anxiety in mice.Methods:The male mice were randomly divided into 7 groups,respectively,the model group,the high,medium,low doses of diazepam group,salvia miltiorrhiza and diazepam in high,medium,low dose group.The model group was given intragastric administration of 0.5%CMC-Na and other groups were treated with intragastric administration of the corresponding drugs,continuous intragastric administration for 5 days.Half an hour after the last administration,recorded the mice open-arms entries accounting for the percentage of the total entry arms numbers(OE%)and open-arms time accounting for the percentage of the total entry arms time(OT%).The numbers of mice passing the box in 10 minutes were recorded by the light-dark box experiment.The intervention effect of the combined application of salvia miltiorrhiza and diazepam on anxiety in mice was observed,and the content of γ-aminobutyric acid,glutamic acid and 5-HT neurotransmitter in brain of mouse were determined by enzyme-linked immunosorbent assay method.Results:The high-dose group of the combination of salvia miltiorrhiza and diazepam could significantly increase the percentage of stay numbers and time entering open arms in the elevated plus maze test,showing the anti-anxiety effects on mice.And the medium dose group could significantly increase the percentage of stay time entering open arms in the elevated plus maze test and the passing times in the light-dark transitions test,showing a synergistic trend of the combined application of salvia miltiorrhiza and diazepam.Conclusion:The combination of salvia miltiorrhiza and diazepam has a trend of increasing the anti-anxiety effect,and its anti-anxiety mechanism may be related to reduce the GLu content of mouse brain.
