摘  要:为探讨鸡肝脏基础型脂肪酸结合蛋白(Lb-FABP)和肝脏脂肪酸结合蛋白(L-FABP)蛋白的配基结合属性,诱导表达并利用Glutathione Sepharose 4B亲和层析纯化了鸡的GST/Lb-FABP和GST/L-FABP融合蛋白,得到的GST/Lb-FABP和GST/L-FABP融合蛋白大小分别为38ku和40ku。将鸡的Lb-FABP和L-FABP分别与1,8-ANS探针结合,结果显示Lb-FABP和L-FABP均表现出对荧光探针的较强结合能力;为进一步测定棕榈酸、花生四烯酸、油酸、胆汁酸和维甲酸这5种配体与Lb-FABP和L-FABP结合的特异性和亲合力,使用这些脂质分子与1,8-ANS探针进行置换分析,结果表明鸡Lb-FABP与维甲酸结合能力最强,而L-FABP与油酸的结合能力最强。本研究结果为深入探究Lb-FABP和L-FABP的生物学功能提供了良好的基础。The aim of this study was to explore the ligand binding property of chicken liver basic fatty acid binding protein (Lb-FABP) and liver fatty acid binding protein (L-FABP). GST/Lb-FABP and GST/L-FABP were induced and then purified by Glutathione Sepharose 4B,which were 38 ku and 40 ku of molecular weight,respectively. The fluorescent probe 1-anilinonaphthalene 8-sulfonic acid (1, 8-ANS) was used to investigate binding property of Lb-FABP and L-FABP. The results showed that both of Lb-FABP and L-FABP displayed high binding ability. In order to quautitate binding specificity and affinity of palmitie acid, arachidomic acid, oleic acid, cholie acid and retinoid acid with Lb-FABP and L-FABP, a competition binding assay was developed to monitor the ability of these lipid molecules to displace bound 1,8-ANS from the binding cavity. The binding to retinoid acid and L-FABP binding to oleic of the biological function of Lb-FABP and L-FABP at results revealed that there were the biggest activity of Lb-FABP acid. This study provides a good foundation for the investigation protein level.
