强精煎对实验大鼠附睾精子抗氧化作用的研究

摘　　要：目的研究以健脾益肾为主要功效的中药强精煎对弱精子症大鼠附睾精子的抗氧化作用。方法取60只雄性SD大鼠随机分4组。给药方法:正常组每天给予生理盐水灌胃;模型组每天给予奥硝唑(ORN)400mg/kg体重灌胃;黄精赞育胶囊组每天给予[ORN(400 mg/kg)+黄精赞育胶囊(200 mg/kg)]灌胃;强精煎组每天给予[ORN(400 mg/kg)+强精煎配方颗粒(10 g/kg)]灌胃。4周后检测大鼠附睾精子的浓度、活动率、附睾匀浆丙二醛(MDA)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)浓度;取睾丸附睾作常规病理分析,观察奥硝唑对大鼠生精功能的影响。结果模型组大鼠附睾精子活动率明显下降,附睾匀浆MDA浓度显著升高,而SOD、GSH-Px浓度明显降低,与正常组相比均有显著差异(P<0.05);强精煎组及黄精赞育胶囊组大鼠附睾精子活动率和MDA、SOD及GSH-Px浓度与正常组比较,无明显差异(P>0.05)。各组大鼠附睾精子浓度无显著性差异(P>0.05)。各组大鼠睾丸生精上皮及附睾上皮完整,生精小管管腔和附睾管腔内精子数量无明显差异。结论中药强精煎可通过增强抗氧化酶活性而发挥抗氧化作用,保护附睾精子免受氧化损伤,保持正常的精子活力。Objective To observe the Antioxidant effect of Qiangjing Decoction which has the function of tonifying the spleen and the kidneys on the sperm of epididymis in rat with asthenospermia. MethodsRandomly divide 60 healthy male rats into normal group,model group,Qiangjing Decoction group and HuangJingzanyu capsule group（n=15）.Delivery method：normal group：normal saline given daily;model group：ORN（400mg/kg body weight） given daily;HuangJingzanyu capsule group：ORN（400 mg/kg） ＋ Huang Jingzanyu capsule（200 mg/kg） given daily;Qiangjing Decoction group：ORN（40 0mg/kg） ＋ Qiangjing Decoction granules（10 g/kg） given daily.Four weeks later,we test the rats’ Sperm concentration,motility,MDA,SOD and GSH-Px concentration.We also take the testis epididymis for routine pathological analysis to learn how ORN affects the spermatogenic function of rats. Resultsthe sperm vitality and the SOD、GSH-Px concentration of model group is significantly lower than the normal group（P＆lt;0.05）;the MDA concentration of model group is significantly higher（P＆lt;0.05）,while no significant difference is shown amount the Qiangjing Decoction group,the HuangJingzanyu capsule group（P＆gt;0.05）.All Rats’ testis seminiferous epithelium and epididymal epithelium is complete,and no significant difference is shown in the sperm counts of Seminiferous tubule lumen and the epididymal lumen. ConclusionQiangjing Decoction can have antioxidant effect by enhancing antioxidant enzyme activity,it can also protect the epidermal sperm from oxidative damage,and maintain the normal sperm motility.
