摘 要:目的研究桂郁金水煎液对离体家兔胸主动脉条的作用,并探讨其作用机制。方法采用家兔离体胸主动脉环灌流模型,制备离体动脉条标本,将离体标本随机分为7组:去内皮组、内皮完整组、溶媒对照组、桂郁金对照组、氯沙坦(10-6mol/L)组、普萘洛尔(10-6mol/L)组、维拉帕米(10-6mol/L)组。去内皮组和内皮完整组分别累积加入桂郁金水煎液使终浓度分别为5.58,11.16,22.32,44.64,89.28 mg/ml,溶媒对照组累积加入等量的纯净水,桂郁金对照组加入桂郁金水煎液使终浓度为44.64 mg/ml,氯沙坦组、普萘洛尔组、维拉帕米组分别用相应的阻断剂孵育10 min后加入桂郁金水煎液使终浓度为44.64mg/ml,观察桂郁金水煎液对离体家兔胸主动脉条收缩张力的作用,以及各阻断剂对此作用的影响。结果桂郁金水煎液达到一定浓度时可明显收缩家兔胸主动脉条,去内皮组与内皮完整组都呈现明显的量效关系,但去内皮组与内皮完整组组间差异不显著;药物溶媒则对动脉条张力变化无明显影响;与桂郁金对照组相比,氯沙坦组和维拉帕米组均有差异显著性(P<0.01),普萘洛尔组则无差异显著性(P>0.05)。结论桂郁金水煎液对家兔胸主动脉条有非内皮依赖性收缩作用,其作用机制可能与AT受体、Ca2+通道有关,则与β受体可能无关。Objective To study the mechanism of Guiyujin water extract on aorta contraction in rabbits in vitro. MethodsUse the thoracic aorta ring perfusion model in rabbits,and divided them into seven groups randomly: the endothelium removal group,the endothelium group,the solvent control group,the GYJ control group,the Losartan group,the Pro group and the Ver group.For the endothelium removal and endothelium groups,GYJ were added accumulately every 15 minutes andmade the final concentrations respectively at 5.58,11.16,22.32,44.64,89.28 mg/ml,for the solvent control group,the same amount of pure water was added;for the GYJ control,losartan,propranolol,verapamil groups,used with corresponding blockers to incubate for 10 minutes(except GYJ control group),then joined GYJ to make the final concentration at 44.64 mg/ml,observed the changes of vascular tension. ResultsGYJ could obviously contract thoracic aorta rings at a certain concentration,the endothelium removal group and the endothelium group had a concentration-response relationship,but no significant difference between the two groups;the solvent had no obviously effect on the tension of thoracic aorta rings;each blockers except the Pro group outside,the other groups had significant difference compared with the GYJ group(P&lt;0.01). ConclusionsThe shrinkage effect of GYJ for the thoracic aorta rings in rabbits was independent of endothelium,the action mechanisms maybe connected with AT receptor、Ca2+ channel,but perhaps had no relationship with β receptor.
