摘  要:目的观察不同浓度人参皂苷Rb1对胎鼠背根神经节(DRG)神经元生长状态及P物质(SP)表达的影响。方法选择胎龄为15天的SD大鼠,获取DRG神经元并进行体外分散培养,48 h后,随机分为对照组、低浓度Rb1(10μg/ml)组、中浓度Rb1(100μg/ml)组、高浓度Rb1(1 000μg/ml)组,继续培养12 h,倒置相差显微镜观察各组神经元的生长状态,免疫组织化学方法检测SP在各组中的表达。结果对照组DRG神经元细胞贴壁呈单层散在分布,突起互相交织形成网状。不同Rb1添加组DRG神经元细胞部分呈簇状聚集,突起较长且相互交织。免疫组化结果显示SP主要表达于DRG细胞质中,添加Rb1后,SP的表达明显多于正常对照组,比较有统计学意义(P<0.05)。随着Rb1浓度的增加,SP表达量没有明显差异。结论人参皂苷Rb1可以促进胎鼠DRG神经元的生长,且能够调节DRG神经元SP的表达,对胎鼠背根神经节神经元具有一定的保护作用。Objective To observe the effect of ginsenoside Rb1 on the expression of SP and morphological changes in dorsal root ganglion neurons. MethodsThe dorsal root ganglion neurons of SD rat embryoes were cultured for 48h,then all cultured cells were divided into 4 groups randomly.In the control group,nothing was added to the cultures;in the low Rb1 group,additional Rb1(10 ug/ml) were added;in the middle Rb1 group,additional Rb1(100 ug/ml) were added,in the high Rb1 group,additional Rb1(1 000 ug/ml) were added.After culture,the morphological changes of neurons in each group were observed.And the expression of SP was detected. ResultsIn the control group,the DRG neurons scattered in a single layer,a little part gathered together.In the Rb1 group,some neurons gathered together and others singled,neurite networks exist and the neurite was longer than normal group and cross-cutting a mesh.Immunohistology result showed that the the expression of SP in Rb1 groups was higher than that of the control group. ConclusionGinsenoside Rb1 can affect the morphological changes and the expression of SP in dorsal root ganglion neurons,it has neuroprotective effect on the neurons.
