摘 要:目的:探讨针刺对血管性痴呆(VD)大鼠血清TNF-α、IL-1β及学习记忆能力的影响,为临床治疗血管性痴呆提供依据。方法:将168只SD雄性大鼠随机分为5组,即:正常组、假手术组、模型组、手针组、电针组。采用四血管阻断法制作VD大鼠模型。造模成功后,进行针刺四神聪、风池穴干预。从不同时间点通过Morris水迷宫测试VD大鼠的学习记忆能力;采用酶联免疫吸附法检测大鼠血清TNF-α、IL-1β含量的变化。结果:同一时间点,手针组、电针组与模型组比较,大鼠逃避潜伏期明显缩短,穿越平台次数增多,差异均具有统计学意义(P<0.05或P<0.01);同一时间点,电针组较手针组穿越平台次数明显增多,差异有统计学意义(P<0.05)。同一时间点,手针组、电针组大鼠血清TNF-α和IL-1β含量与模型组比较明显降低,差异具有统计学意义(P<0.01)。结论:针刺四神聪、风池穴能改善VD大鼠的学习记忆能力,具有脑保护作用,能降低VD大鼠血清中的TNF-α、IL-1β含量,并且电针比手针效果显著。Objective: To explore the effects of acupuncture on the levels of TNF-α and IL-1β in serum and on the ability of learning and memory in vascular dementia (VD) rats, to provide the basis for the clinical treat- ment of vascular dementia. Methods: 168 male SD rats were randomly divided into five groups, including nor- mal group, sham operation group, model group, acupuncture group and electro - acupuncture group. VD rat model was made with the four - vessel occlusion method. After successful modeling and Fengchi points, to test learning and memory in rats at different time points by changes , acupuncture on Sishencong Morris water maze ; to detect of TNF-α,IL-1β content levels in serum with the enzyme- linked immunosorbent assay. Results: At the same time point, the acupuncture group, EA group compared with model group, the rats escape latency was significantly shorter and the times of over - stepping of platform was increased, and the differences were sta- tistically significant(P 〈0.05 or P 〈0.01 ) ;at the same time point, the times of over- stepping of platform of EA group was significantly increased than those in acupuncture group, and the difference was statistically signifi-cant(P 〈0.05). At the and EA group compared significant( P 〈0.01 ) ; at same time point, TNF-α and IL-1β content levels in serum of acupuncture group with model group were significantly decreased, and the differences were statistically the same time point, TNF - ctcontent levels in serum of EA group puncture group was significantly decreased, and the difference was statistically significant( P 〈 0.05). Conclu- sion: Acupuncture on Sishencong and Fengchi can improve learning and memory ability of VD rats, and it has a protective effect on brain. Moreover, it can also reduce TNF-α and IL-1β contents in serum of VD rats, so e- lectro- acupuncture is more significant.
