摘 要:目的:观察大鼠心脏移植急性排斥反应中的移植心脏存活和心肌细胞的凋亡情况及苏木乙酸乙酯提取物对其影响。方法:以Wistar大鼠为供体,以SD大鼠为受体,建立大鼠腹腔异位心脏移植模型,应用原位末端标记(TUNEL)技术检测移植心脏切片,进行排斥反应,计算凋亡指数(AI)。结果:细胞凋亡主要发生于心肌细胞;苏木乙酸乙酯提取物组和苏木加半量环孢素组都能能明显减少心肌细胞的凋亡,减轻移植心脏的组织损伤。结论:在大鼠心脏移植模型中,苏木乙酸乙酯提取物能明显抑制心肌细胞凋亡,减轻移植心脏的急性排斥反应。Objective:In order to observe the survival condition of the transplanted heart in acute heart transplantation rejection, we want to observe the cardiac muscle cell apoptosis condition in the acute heart transplantation rejection process. We also want to observe the effect of Ethyl Acetate extracted from Sappan Wood on the cardiac muscle cell apoptosis condition in the acute heart transplantation rejection process. Methods:In the experiments for the model of heterotopic cardiac transplantation in rats, Wistar rats were donors and SD rats were recipients. TUNEL assay (Terminal deoxynucleotidyl transferase mediated dUTP Nick End Labeling assay) was used to test the section of the transplanted heart. After heart transplantation rejection, apoptotic index (AI) was obtained. Results : Cells from Sappan Wood or Ethyl Acetate diac muscle cells and alleviate the apoptosis occurred mainly in cardiac muscle cells. Ethyl Acetate extracted added with half ciclosporin could significantly reduce the apoptosis of cardamage of the transplanted heart's tissue. Conclusion:With the experiments for the model of heterotopic cardiac transplantation in rats, Ethyl Acetate extracted from Sappan Wood can significantly inhibit the apoptosis of cardiac muscle cells and alleviate the acute heart transplantation rejection.
