摘 要:目的:观察新加达原饮(IXJDY)体外抗乙型肝炎病毒(HBV)的作用。方法:采用四甲基噻唑蓝(MTT)法检测IXJDY对HepG2.2.15细胞的半数毒性浓度(TC50)和最大无毒浓度(TC0);在TC0基础上应用不同浓度药物作用于HepG2.2.15细胞,分别在第72h和144h收集细胞培养上清液,荧光定量PCR法测定细胞外HBV DNA的含量。结果:TC50=14.22mg/mL,TC0=8.13mg/mL,新加达原饮对HepG2.2.15细胞毒性较低。无毒浓度的新加达原饮在HepG2.2.15细胞培养中可有效地抑制细胞HBV DNA的分泌(P<0.05,P<0.01);且呈现量效和时效关系。结论:IXJDY在体外有显著的抗HBV的作用,且毒性较低。Objective: To study the inhibitory effect of influence of XinjiaDaYuanYin (IXJDY) on HBV in vitro. Methods:The TC50 and TC0 of IXJDY to the HepG2.2.15cells were determined by MTT assay. At non - cellulotoxic concentration, the different concentrations of IXJDY were added respectively to HepG2.2. 15 cells,which Were incubated subsequently. At 72h and 144h after treatment with drugs,the levels of HBV DNA were determined by PCR. Results : TC50 = 14.22mg/mL, TC0 = 8.13mg/mL. IXJDY had little cellulotoxicity. IXJDY at the largest non - eellulotoxie concentration could suppress HBV DNA expression in HepG2. 2.15 cells(P 〈0.05 ,P 〈0.01 ). IXJDY was an effective and low toxic anti -HBV drug. Conclusion:IXJDY can inhibit significantly HBV in vitro, and is a lower toxic drug.
