摘  要:目的分析虫草属分子聚丙烯酰胺凝胶电泳银染检测效果,建立一种用于虫草属的DNA分子指纹高分辨率的聚丙烯酰胺凝胶电泳银染检测方法。方法分别对虫草属分子聚丙烯酰胺凝胶电泳的凝胶终浓度、胶电泳步骤、胶固定方案、胶染色方法、胶显色步骤进行分析,确定利于虫草属分子聚丙烯酰胺凝胶DNA多态性展现的方案。结果确定虫草属分子聚丙烯酰胺凝胶电泳银染检测方案,分别为聚丙烯酰胺凝胶浓度为6%、银染染色液为0.3%的AgNO3、胶显色步骤中显影液为强碱NaOH和0.5%的甲醛溶液,且NaOH的浓度为2%。结论本研究所建立的银染检测及显带体系适用于虫草属基因组聚丙烯酰胺凝胶电泳研究。Objective To analyze Cordyceps molecular polyacrylamide gel electrophoresis and silver staining effect,and establish the method of Cordyceps molecular polyacrylamide gel electrophoresis and silver staining.Methods Methods of polyacrylamide gel electrophoresis and silver staining were used.The final concentration of gel elec trophoresis step,the fixing solution of the gum,gum dyeing method,gum color-step were analyzed especially.Results The final dectction methods were determined.Polyacrylamide gel concentration of 6% and 0.3% AgNO3 concentration were suitable for the Cordyceps molecular detection.At the same time,2% NaOH and 0.5% Formaldehyde solution were allowed.Conclusion This study has established the reaction system which is suitable for Cordyceps genome polyacrylamide gel electrophoresis and silver staining.
