骨髓基质干细胞分泌因子对迟发型超敏反应的影响

摘　　要：目的探讨骨髓基质干细胞(BMSCs)分泌因子对迟发型超敏反应的影响。方法分离培养大鼠BMSCs,贴壁法纯化,收集第三代BMSCs上清液获取干细胞分泌因子。制备小鼠接触性皮炎模型,观察干细胞分泌因子对实验小鼠耳廓肿胀的抑制作用及对胸腺指数和脾脏指数的影响。分离培养小鼠脾细胞,MTT比色法检测干细胞分泌因子对淋巴细胞增殖的影响。结果与模型组相比,干细胞分泌因子可明显减轻接触性皮炎模型小鼠耳廓肿胀程度(F=21.429,t=3.658,P<0.01),降低胸腺指数和脾指数(F=19.829、53.951,t=2.434、3.729,P<0.05),抑制ConA诱导的致敏小鼠脾淋巴细胞增殖(F=14.032,t=2.619,P<0.05)。细胞因子高剂量组对模型小鼠胸腺和脾脏淋巴细胞的抑制作用强于低剂量组(F=22.317、26.247,t=2.302、2.781,P<0.05)。结论BMSCs分泌因子对迟发型超敏反应有抑制作用。Objective To explore the effect of bone marrow stem cells （BMSCs） secrete factors on delayed hypersensiti- vity. Methods Rat BMSCs were isolated and purified by adherence screening. The stem cell secrete factors （SCSFs） were ob- tained from the supernatants of third-generation BMSCs. A model of contact dermatitis in rats was created. The depressant effect of SCSFs on swelling of auricle, and their influence on thymus and spleen index were observed. The spleen cells of rat were isolated and cultured. The effect of SCSFs on lymphocyte proliferation was detected by MTT test. Results Compared with the model group, SCSFs could markedly decrease the swelling of auricle in rats in contact dermatitis group （F = 21.429, t = 3.658, P d0.01）, reduce thymus and spleen index （F=19.829,53.95]t=2.434,3.729;P〈0.05）, and inhibit lymphocyte multiplication induced by ConA （F=14.032 ,t =2.619,P〈0.05）. The depressant effect of high-dose cytokine on thymus and spleen lymphocyte was stronger than that of low-dose one （F=22.317,26.247;t=2.302,2.781;P〈0.05）. Conclusion BMSCs have a depres-sant effect on de- layed hypersensitivity.
